Taihe silky fowl is a kind of Chinese tradaitonal and precious medicine with high food, medicinal and ornamental value, and has been classified as an international standard variety. However, the healthcare function and pharmacological effects of Taihe silky fowl have rarely reported yet. The aim of the present study was to investigate the anti-oxidant effect of SF-M on D-galactose induced aging mice and the anti-fatigue effect of Taihe silky fowl.In this experiment SF-M was extracted from Taihe silky fowl by enzymatic method and hydrochloric acid method, and the optimum extraction conditions of SF-M were determined by single-factor experiments and orthogonal tests of these two methods. The results showed that the optimum extraction condition of SF-M with enzymatic method was obtained as follows:the use of papain, pH 6.0, temperature 65℃, the ratio of meal to liquid 1:2, the ratio of enzyme to substrate4% for 4 hours; the optimum extraction condition of SF-M with hydrochloric acid method was the ratio of meal to liquid 1:4, reflux temperature 95℃, extraction time 2 hours. Besides, extraction ratio, anti-free radical properties, light and heat stabilities of SF-M from two extraction methods were studied and compared. The result showed that the enzymatic extraction method was better than hydrochloric acid extraction method, and the extraction ratio of SF-M with enzymatic extraction was higher than the hydrochloric acid extraction, the anti-free radical properties and the heatstability of SF-M with enzymatic extraction were better than the hydrochloric acid extraction.In order to investigatethe anti-oxidant effect of SF-M,60 male KM mice were randomly and averagely divided into six groups, daily injected 0.5g/kg D-galactose through subcutaneous for making aging mice model, and blank control group was injected saline. At the same time, SF-M group were orally given SF-M at the dosages of 50mg/kg,250mg/kg,1250mg/kg; positive group was orally given 100mg/kg vitamin E; model control group and blank control were orally given distilled water. After 6 weeks, MDA, lipofuscin, SOD, GSH-Px, T-AOC were detected. Results showed that model control group compared with blank control group, contents of MDA of liver tissue and serum and lipofuscin of brain and liver tissues were significantly higher (p<0.01), which showed that modeling was successful; high, medium, low-dose SF-M groups and positive group compared with model control group, contents of MDA of liver tissue and serum and lipofuscin of brain and liver tissues were significantly lower (p<0.01, p<0.05), contents of SOD, GSH-Px, T-AOC of liver tissue and serum were significantly higher (p<0.01, p<0.05). Conclusions:SF-M could reduce lipid per-oxidant content and increase anti-oxidative enzyme activity,has a certainanti-oxidative capacity.In order to investigate the anti-fatigue effect of Taihe silky fowl,120 male KMmice were randomly divided into six groups,high-dose silk fowl soup group (1:1), low-dose silk fowl soup group (1:2), high-dose enzymatic hydrolyzed materials group (5mg/g), low-dose enzymatic hydrolyzed materials group (1mg/g), the positive control group (American Ginseng 300mg/kg) and blank control group (distilled water), each mouse was daily fed different subjects at certain volume (0.4mL). After 6 weeks,10 mice of each group were used for weight-bearing swimming test, and another 10 mice of each group were forced to swim for 30min, then some indicators such as blood glucose, blood lactate, BUN, liver glycogen and muscle glycogen, LDH, CK, SOD, GSH-Px were determined. Results showed that Taihe silk fowl soup and enzymatic hydrolyzed materials significantly prolonged the swimming time, reduced the exercise blood lactate and urea nitrogen levels, increased blood glucose, liver glycogen and muscle glycogen contents, reduced lactate dehydrogenase levels and creatinekinase levels, and increased SOD and GSH-Px levels (P<0.05 or P<0.01).Conclusion:Taihe silk fowl soup and enzymatic hydrolyzed materials has obvious anti-fatigue effects.
