Objective To investigate the effect of cannaboid WIN55,212-2on the expressionof cyclic adenosine monophosphate (cAMP) in bovine trabecular meshwork cells(BTMC) in vitro, then discuss its mechanism of reducing intraocular pressure byincreasing aqueous humor eduction.Methods BTMC were primary cultured and subcultured in vitro,immunohistochemical method were used to identity BTMC. The levels of cAMP insupernatants were measured by radioimmunoassay when BTMC were stimulated bydifferent concertrations of WIN55,212-2.Results When BTMC were stimulated by WIN55,212-2of concentrations within0.1~20mol/L,the levels of cAMP in supernatants significantly increased, but whenthe concentration further increased (40-60mol/L),they were no longer rised.Conclusion WIN55,212-2may play the role of reducing intraocular pressure byincreasing the levels of cAMP in BTMC.
