Objective:To investigate the infects and mechanisms of Tanreqing injection on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in ratMethods:56 SD rats were randomly divided into 7 groups:normal control group; LPS model of 2h,4h,6h group; Tanreqing treatment of 2h,4h,6h group. LPS (5mg·kg-1) was injected into tail vein in LPS model group and Tanreqing treatment group, 1mL Tanreqing injection was injected into another tain veil in Tanreqing treatment group. As control, lml sterile saline was injected into tail vein in normal control group. The rats were killed at each observation time point. The pathological changes of lung were observed, and lung wet/dry ratio (W/D), the ratio of polymorphonuclear neutrophil (PMN%) in bronchoalveolar lavage fluid (BALF), Pulmonary permeability index (LPI) were calculated.In addition, the concentrations of nitric oxide (NO) and malondialdehyde (MDA), the activities of superoxide dismutase(SOD) and glutathione peroxidase (GSH-Px) in lung tissues were measured by the method of colorimetry. The expression of NF-κB P65 in lung tissue were detected with immunohistochemisty,Results:The acute lung injury model was successfully induced by LPS. Compared with the control group, The pathological changes of lung in LPS model group were significant; PMN%, lung W/D, LPI and MDA, NO of lung tissue were also significantly increased (P <0.05); SOD and GSH-px in lung tissue were significantly decreased (P<0.05). Compared with LPS model group, PMN%, lung W/D, LPI inTanreqing treatment group were significantly decreased(P<0.05)at each observation time point, and MDA, NO of lung tissue were decreased significantly (P<0.01) at 4,6 h, while the lung tissue SOD, GSH-px activity were significantly increased at 4,6 h (P<0.01). Compared with the control group.the expression of NF-kB P65 in lung tissue was significantly up-regulated in LPS model group.Compared with LPS model group, the expression of NF-κB P65 in lung tissue in Tanreqing treatment group were significantly down-regulated(P<0.01)at 4,6 h.Conclusion:Tanreqing Injection shows a protective effect on LPS-induced acute lung injury rats.The mechanism may be related to its ability of clearing the reactive oxygen species, inhibiting the activation of the signal pathway of NF-κB and inflammatory cascade...
