Objective:Patients by in vitro synthesis to Newcastle disease virus to new inspection, this new type of virus observed the destructive effects on cancer cells, Newcastle disease virus is treated in the integration of the pulsed dendritic cells (DC) and cytokines Induced killer cells (CIK) generated on the basis of two new viruses (our group has completed the part of the experiment, Shandong Provincial Natural Science Foundation of China, Project Fund No.:Y2006C22), Newcastle disease virus on the destruction of key test cancer cells The practical effect of the immune antigen.Methods:Get Qualified peripheral blood, extracted from peripheral mononuclear cells exist, from pulling out of DC, CIK cells. DC sensitized on which dealt with by the SGC-7901 gastric cancer cells were sensitized activities. CIK then normal processing with pulsed DC after fusion culture, cultivate treated DC induced by variation of CIK cells (antigen-DC-DIK). To antigen-DC-CIK cells for the effect to the role by NDV SGC-7901 cells and the role without NDV cells SGC-7901 target cells by MTT cytotoxicity assay, target cells were observed after the killing effect of blocking MHC molecules Changes.Results:At 20:1 effector-target ratio in vitro, the cytotoxic activity of antigen-DC-CIK against SGC-7901 which ware modified or not modified by NDV was (89.0±1.4)% and (69.3±1.5)% respectively (P<0.01); After target cells'MHC molecule was blocked, the cytotoxic activity was reducd by 30.8% and 21.1% respectively. This indicates specific cytotoxic activity was enhanced apparently after antigen was modified by NDV.Conclusions:The conclusion of this experiment:The experiment proved that the same antigen-DC-CIK can NDV shared the role of cancer cells, is a new way of biological therapy, can effectively deal with gastric cancer cells, cancer cells can enhance the effect The role of antigen, NDV antigen in gastric cancer cells by enhancing the antigen to enhance immune response to tumor cells. With this treatment can make the immune cells to destroy cancer cells remained at 9.7% efficiency or so.
