Objective:To evaluate the clinical significance of detecting K-Ras gene point mutation and CA19-9, CA242 test in peripheral blood of patients with pancreatic carcinoma.Methods:In the same period 12 patients with pancreatic carcinoma and 17 non-pancreatic carcinoma, we studied K-ras gene 12 point mutation by PCR-MASA (mutant allele specific amplification) and detected CA19-9, CA242 by ECLIA (Electro-chemiluminescence Immunoassay) in peripheral blood. The data were analysed with X2 test and Fisher's exact test. Results:The rate of K-ras gene 12 point mutation in pancreatic carcinoma group was 75%(9/12),in non-pancreatic carcinoma group the rate was 5.88%(1/17); The sensitivity and specificity of K-ras gene mutation in diagnosis of pancreatic carcinoma were 75.00% and 94.12%; combining CA19-9 with K-ras gene mutation in parallel experiments of pancreatic cancer can improve the diagnostic sensitivity to 83.33%. there is no correlation between CA19,CA242,k-ras gene mutation and the patient's sex, age, tumor size, tumor location and TNM stage.(P> 0.05). Conclusions:(1)The diagnostic specificity of joint detection of CA19-9 and K-ras gene mutation in peripheral blood of patients is higher than that of single detection of k-ras gene mutation in pancreatic carcinoma, and which is conducive to the diagnosis of pancreatic carcinoma.(2) There is no correlation between CA19-9,CA242,k-ras gene mutation and the patient's sex, age, tumor size, tumor location and TNM stages in pancreatic carcinoma group. (3) Elevated CA242 in peripheral blood has not significantly statistically significant between in pancreatic carcinoma group and in non-pancreatic carcinoma group,further reacher is needed.(4) PCR-MASA (mutant allele-specific amplification) simple,rapid, specific and sensitive.The amplification products can be observed only by conventional electrophoresis and staining. Restriction enzyme digestion, hybridization, radioactive and non-radioactive imaging are not needed.
