4-Cumylphenol is organic peroxide that is widely used in the petrol-chemistry, plastics and new materials fields. In recent years, with the increased use, 4-Cumylphenol can be detected from the water body, drinking water and the soil. 4-Cumylphenol has been detected in raw water and finished water of Shanghai Huangpu River. Current the standards for drinking water quality do not include 4-Cumylphenol, therefore, the information on previous surveys and research are scare. Owing to lack of the basic toxicological data and exposure information on human, there is no standards for 4-Cumylphenol. Moreover, there are no methods for the detection of 4-Cumylphenol. The investigation shows that the current commonly used conventional water treatment processes can not be effective removed 4-Cumylphenol in raw water. Thus, the population are inevitable expose to 4-Cumylphenol from drinking water in long-term life.In this study, the qualitative and quantitative detection methods of 4-Cumylphenol were established, acute test was employed to get LD50 of 4-Cumylphenol, Ames test and micronucleus test were used for the detection of the genetic toxicity of 4-Cumylphenol. These results will provide basic data to the study of drinking water standards on 4-Cumylphenol.Part I The detection methods for 4-Cumylphenol in drinking waterSolid phase extraction was used to enrich 4-Cumylphenol in drinking water, the concentration of 4-Cumylphenol in different water was detected by gas mass spectrometry (GC/MS). The experiments show that the established methods has a good seperated effect and the ideal linear relationship.The minimum detection limit is up to 0.02μg/L, recovery rate is 82.1%~86.2%, precision is 6.8%-14.8%. This method can be used for qualitative and quantitative analysis of 4-Cumylphenol in different water.The results also show that the 4-Cumylphenol can be detected in raw water and finished water from Huangpu River, the concentration of 4-Cumylphenol were 0.35μg/L and 0.22μg/L, respectively. The current conventional treatment technology can not effectively remove 4-Cumylphenol.Part II LD50 of 4-CumylphenolLD50 of 4-Cumylphenol was tested by oral acute toxicity test.The result show that 4-Cumylphenol can lead to weight loss of animal in high dose group and cause pathological changes in cell proliferation in glomerular. LD50 for mice is 1141.5mg/kg by oral administration.95% confidence interval is 744.75-1749.62mg/kg. According to Lawton's toxicity grading criteria belongs to no toxic substance.Part III The study of the genetic toxicity on 4-CumylphenolUsing Salmonella typhimurium reverse mutation assay and micronucleus assay in bone marrow polychromatic erythrocytes were employed to test the genetic toxicity of 4-Cumylphenol from the levels of gene and he of chromosome, respectively. The results show that in all dose groups, regardless of with or without S9, there is no significant increase in reverse mutation of Salmonella typhimurium histidine auxotrophic mutant strain both in TA97 and TA98. These results indicated that 4-Cumylphenol can not cause frame shift mutation and base substitution mutation.Micronucleus test showed that micronucleus rate in bone marrow polychromatic erythrocytes of mice is no significant difference in different doses of 4-Cumylphenol compared with the control group (p>0.05).This results show that current dose of 4-Cumylphenol can not induce mouse bone marrow micronucleus rate increased and suggested that 4-Cumylphenol is not a broken agent of centric. |
