| Objective:The study was aimed to observe the protective effects of lipoxin A4 on acute necrotizing pancreatitis associated lung injury in rats.Methods:Seventy-two SD rats were randomly divided into three groups(N=24): sham operation group(S group), acute necrotizing pancreatitis group(ANP group), treatment with lipoxin A4 group(Tgroup). The model of ANP was induced by the retrograde injection of 5% sodium taurocholate in the pancreatobiliary duct. Then put rats to death after operation at 12h, 24h. Serum level of amylase , TNF-α, IL-1β, IL-10, E-selectin were detected at 12h, 24h time points.The activity of MPO in pancreastic and pulmonary tissues were also measured. the pathological changs of lung and pancreas were observed under microscope.Results:1,The levels of serum AMY, TNF-α, IL-1β, E-selectin in ANP group were remarkably higher than those in S group at 12h, 24h time points (P<0.05).The activity of MPO in pancreastic and pulmonary tissues and the pathological scores of pancreas and lung were also remarkably higher in ANP group than those in S group at the same time points(P<0.05). Compared with ANP groupe ,The levels of serum AMY, TNF-α, IL-1β, E-selectin , The activity of MPO in pancreastic and pulmonary tissues, and the pathological scores of pancreas and lung were remarkably reduced in T group during the same time points(P<0.05).2,serum levels of IL-10 in ANP group were significantly higher than that in S group at 12h, 24h time pionts(P<0.05), but significantly lower than that inT group at12h,24h time points (P<0.05). Conclusion:1,The method with which the models of ANP were established by retrograde injection of 5% sodium taurocholate into the biliopancreafic duct is useful and duplicated.2,Lipoxin A4 could exert protective effects on acute pancreatitis associated lung injury in rats with ANP by pulmonary histological improvement. The mechanisms probably is related to the decreased level of neutrophil infiltration in pulmonary tissue, production of pro-inflammatory cytokines and adhesion molecule, and the increased level of anti-inflammatory cytokines. |
