| "Jianganle keli"in the clinication for years and has already listed sales of famous proprietary Chinese medicine at present.It has raise blood to protect liver,detoxification and relive pain,reduce transaminase,subsidise jaundice and improving all kinds of hepatitis clinical symptoms role."Ganduqing pill"is the secondary investigative product based on"Jianganle keli"."Jianganle keli"root in"shaoyao gancao decoction"that recorded in Zhangzhongjing′s"shang han lun",it is a king of proprietary Chinese medicine for treatment of hepatitis ,it′s bases effective component is extracted from White Paeony Root and Liquorice.Aim to provide scientific evidences for develop"Ganduqing pill", This test studied it′s extraction and purification technology,preparation procedure and pharmacology activity.the main research works carried out are as follows:1. established high performance liquid chromatography (HPLC) assay, with paeoniflorin,glycyrrhizic acid and liquiritin as investigates index ,combine"Jianganle keli"extraction and purification technology alter it′s parameter to optimize the technology of GanDuQing pills.The optimized result is as follows:the liquorice particlesize is 0.9-2mm,white Paeony Root is 0.9-2mm,ratio of material-fluid is 1:16 and soaking time is 3h,extraction 3 times and each time is 2h,relative density of soup is 0.75g/ml,sediment 1 times and stewing 48h,on this technological condition,the total extraction rate of"GanDuQing pills"is 15.44mg/g. But the total rate of"Jianganle keli"is 12.68mg/g.It follows that the content of active principle has improved.2. investigate and optimize the processing technology of"GanDuQing pills",the respectively from ground substance,ratio of drug and group prorata,temperature of medication solution,distance and speed of drip,kind of coolant,etal.The optimized project was as follows:simethicone as coolant,the ratio of medicine and matrixis 1:5,PEG4000 and PEG6000 (10%beewax)is 1:3,the distance of dropping is 7cm,the temperature of medication is 90oC,the dropping speed is 40～50d/min.On this condition that the appearance quality of pills,weight variation and disintergration time limited all accord with the request of pharmacopeia.3. Investigate and contrast the effect of anti-inflammatory,analgesia and liver injury of"GanDuQing pills"and"Jianganle keli", in the experiment of anti-inflammatory, we use two indexes:one is ear swelling degree by dimethylbenzene,determine mouse ear edema and it′s inhibition rate;the other is the capillary permeability in abdominal cavity by the acetic acid,absord peritoneal fluid and determine the number of OD. in the experiment of analgesic,also have two indexes:one is hot-plate,calculate the threshold;the other is the writhing reaction by dimethybenzene,count the degree of torsive body of the mice.Above index all arranged eight groups. Moreover, the liver injure by CCl4 induced experiment have nine groups and two indeies:one is GPT content in rat blood,the other is GOT. The result show that if based on the same born dosage,"GanDuQing pills"is superrior to"Jianganle keli"on anti-inflammation,abirritation and liver injury. |
