Objective:Study the role of p38MAPK in the signal pathway of MAPK regulating the expression of IL-4 and IL-5 and its dependablity by peripheral blood mononuclear cell in allergic rhinitis.Methods:Selected 40 patients with acute episode of allergic rhinitis and divided randomly into two groups:control group, experimental group,20 cases each.Selected 20 normal persons as the control group. Each case was drawed blood from ulnar vein.Cultured peripheral blood mononuclear cells from venous blood in 24-well plates in which the experimental group was stimulated with 40μg/ml SB203580, the blank group and control group was stimulated with the same volume of DMSO (solvent of SB203580)for 4 hours.The expression of p38MAPK were observed by Western blot,.The expression of IL-4 and IL-5 mRNA were observed by RT-PCR,IL-4 and IL-5 protein in supernatants were measured by ELISA.Results:The expression of nuclear p38MAPK protein,IL-4 and IL-5 mRNA,IL-4 and IL-5 protein in supernatants of allergic rhinitis PBMC inhibited by SB203580 were significantly lower than those of allergic rhinitis PBMC stimulated without SB203580(P=0.00<0.05, respectively).There were good positive correlation between the p38MAPK and the expression of IL-4 and IL-5 mRNA(P=0.00<0.05),and also good positive correlation between the p38MAPK protein and IL-4 and IL-5 protein in supernatants (P=0.00<0.05).Conclusion:The inhibitor SB203580 of allergic rhinitis PBMC inbihited p38MAPK and down-regulated expression of the level of reverse transcription and protein of IL-4 and IL-5.The activation of p38MAPK signal pathway of PBMC participated in pathogenesis of allergic rhinitis. |
