Objective: To observe the effects of high glucose on chemokine ligand 16 (CXCL16) expression and secretion in THP-1 macrophages and intervention of aspirin in the activity.Methods:(1) Macrophages was exposed to medium containing 33.6 mmol/L glucose for different time(0,6,12,24,48,72 and 96h);(2) Macrophages was exposed to medium containing glouse at different concentration (5.6,11.2,16.8 and 33.6 mmol/L)for 72 hours;(3) Macrophages was exposed to medium containing 33.6 mmol/L glucose and different concentration of asipirin(0,12.5,25 and 50 ug/ml) for 72 hours.RT-PCR and ELISA was used to detect the CXCL16 mRNA expression and protein secretion respectively.Results:(1) With the extension of time, as well as the elevated glucose concentration, CXCL16 expressed and secreted by macrophages increase and reach the peak at 72 hours.(2) With the glucose concentration of medium increases, macrophages expression and secretion CXCL16 increase accordingly.(3) With the medium of aspirin,concentration of CXCL16 expressed and secreted by macrophage decreased while the concentration of asipirin in the medium increased. Conclusion: High glucose can up-regulate the expression and secretion of CXCL16 in THP-1 macrophages ,however aspirin can inhibit the activity significantly.
