| Objective: Upon the inhibition effect that RGDS (comes from fiber adhesion protein fN, short chain repeat sequence biological active peptide, arginine- glycine– aspartic-acid serine (RGDS) sequence )on mice liver metastases after those spleens were inoculated with CT26 cells , discusses the inhibition mechanism, and the application prospect of prospects do a preliminary study.Methods: First, established liver metastatic carcinoma animal model, by CT26 cells vaccinated into mice, spleen (control group). 0.2 ml (concentration for 1 x 106 / mL) CT26 cells levitation liquid was injected into those spleens (experimental group), determine the liver metastases model rate. Based on that model, mice, tail with bioactive peptide RGDS intravenous. Observe the inhibitory effect on liver metastasis that RGDS does upon CT26 cells mice spleen vaccination model (A group to C group) and the difference of the survival time between the two groups (B group to D group).Results: Mice, spleens after 0.2 mL (1 x 106 / mL) CT26 cells injection groups, there were 19 mice and 20 mice liver, respectively, found metastatic tumor nodules. Accordingly, their liver metastasis rate is 95% and 100%. The two experimental groups, there were 11 mice and 9 mice liver, respectively, found metastatic tumor nodules. Accordingly, their liver metastasis rate is 55% and 45%.Experimental group C compared with control group A, their liver metastasis rates were significantly different (p < 0.05), with a statistical significance. Experimental group D compared with control group B, their liver metastases rates were significantly different (p < 0.05), with astatistical significance. And the surviva time of mice in Experimental group D was longer than mice in group B.Conclusion: Liver metastatic carcinoma animal model, established by CT26 cells vaccinated into Mice, spleen, is a mature colorectal cancer liver metastases model, and it's transfer rate is higher. Biological active peptide RDGS has a significant inhibitory effect on CT26 cells, liver metastasis. The mechanism is that RDGS can compete ECM and fibrin with the integrin molecular, which is on the surface of tumor cells and platelet, interferes tumor cells - ECM interaction, inhibit the form of cancer embolus, so as to achieve the purpose of inhibiting tumor metastasis. |
