Objective: Through the observation of the influence of neurotransmitter of Prostaglandin E1 to the phontype and function activity of macrophage in the uterus of normal mice, to explore its mechanism of immune suppression and the relationship between maternal-fetal immune tolerance.Methods:Non-pregnant normal Kunming mice were randomly divided into control group (A group) and three experimental groups E-SG, E-SZ, E-SD. Experimental group were intravenous 1303ng/kg/d, 2606ng/kg/d, 3909ng/kg /d of prostaglandin E(10.2ml),with a sterile saline(0.2ml) control group, continuous injection of 5 days. After the last injection, respectively 1h, 3h, 6h, 12h, 36h to take the uterus. Frozen sections and paraffin sections made and used non-specific esterase staining and immunohistochemical method to detec the phenotype and functional activity of uterine macrophages.Results:(1)Comparing with control group mice, After the injection of PGE1for 1h, the expression of uterus macrophage CD14+ at the endometrium, myometrium and perimrtrium of every experimental group showed an apparent down-regulation (P<0.01). After the injection of PGE1 for 3h, the expression of uterus macrophage CD14+ at the endometrium and perimrtrium of every experimental group showed an apparent down-regulation (P<0.01),the expression of macrophage CD14+ at the e perimrtrium of every experimental group showed an apparent increase. (P<0.01). After the injection of PGE1for 6h, the expression of uterus macrophage CD14+ at the endometrium of every experimental group showed an apparent down-regulation (P<0.01).and the expression of uterus macrophage CD14+ at the perimrtrium of every experimental group showed an down-regulation (P<0.05).(2)There is no macrophage CD204+ found in control group and low concentration group. They only been found in medium experimental group after the injection of PGE1 for 12 hour and 36 hour. They also been found in high experimental group after the injection of PGE1 for 6 hour, 12 hour and 36 hour. CD14+ and CD204+ had a similar change tendency. The high experimental group after the injection PGE1 for 36 hour had significant increase. (P<0.01).(3)Comparing with control group mice, After the injection of PGE1 for 1h, the expression of uterus macrophageα-NAE+ at the endometrium of every experimental group showed an significant chage,Macrophageα-NAE+ at the perimrtrium had significant increase . After the injection of PGE1 for 12h, the expression of uterus macrophageα-NAE+ at the endometrium of every experimental group showed an significant increase,Macrophageα-NAE+ at the myometrium had significant decrease .After the injection for 36h, the expression of uterus macrophageα-NAE+ at the endometrium, muscularis and outer membrane of high experimental group showed an apparent increase. Conclusion:(1)PGE1 can inhibit the migration and activation of macrophage ,promote the migrate from the endometrium to the perimrtrium,and this effect within a certain range was positively correlated with the dose and half-life related. As time goes by, PGE1 can promote he migration and activation of macrophage.(2)A certain concentration of PGE1 can promote the expression of uterine macrophage receptor CD14+ and CD204+ .(3)PGE1 regulate the activity and function of macrophage by the way of adjusting their amount and acceptors of CD14+ and CD204+ to take part in the occurrence and maintenance of uterine immune tolerance.
