Objective To compare acute toxicity of Wan-Sheng-Hua-Feng-Dan (WSHFD), cinnabar, realgar, arsenite and mercuric chloride (HgCl2) in mice.Methods Mice were given a single gavage of WSHFD 3 g.kg-1 (contains 0%,3% or 10% cinnabar and realgar),0.3 g.kg-1 of ralgar and cinnabar,0.036 g.kg-1 NaAsO2 (10% As of realgar) and 0.07 g.kg-1 HgCl2 (20% Hg of cinnabar). Eight hours later, tissue As and Hg contents, blood biochemistry, histopathology, and metal toxicity biomarker metallothionein expression were determined. Results Accumulation of As after arsenite and Hg after HgCl2 was much higher than other groups. Increased ALT was seen in arsenite-treated animals, while elevated UREA was observed in the HgCl2 group. Histology showed more lesions in arsenite and HgCl2 groups, despite much lower As and Hg doses were used. RT-PCR analysis indicated that the expression of MT-1 increased 4-10 folds in liver and kidney of arsenite and HgCl2 groups, but not in other groups. Conclusions Acute toxicity of cinnabar, realgar, and WSHFD was much less than arsenite and HgCl2, and the use of total As and Hg contents to evaluate the safety of cinnabar- and realgar-containing traditional medicines is inappropriate. Objective:To study the effects of Wan-Sheng-Hua-Feng-Dan on cerebral ischemia in rats. Methods:Adult SD rats were orally givern for four days with the original Wan-Sheng-Hua-Feng-Dan prescription 1.5 g·kg-1, An-Gong-Niu-Huang-Wan 1.5 g·kg-1,Brain injury was produced by the suture method; ischemia for 1 hours,then reperfusion for 72 h, with continous drug administration the brain infarction areas were examined by infarction rate, the mortality in rat, Brain As and Hg content was determined by AAS. Results:the rate of brain infarction was significantly reduced after treatmeat with Wan-Sheng-Hua-Feng-Dan and An-Gong-Niu-Huang-Wan. And brain As and Hg Content was elevated. Conclusion:Wan-Sheng-Hua-Feng-Dan and An-Gong-Niu-Huang-Wan are effective against cerebral ischemia and reperfusion in rats.,and the Protection appears to be due,at least in part,to incrrased As and Hg Content in the Brain.
