Objective:This paper mainly studies the expression of aquaporins (AQPs) in fetal membranes and placenta of gravida with normal pregnancy and oligohydramnios, understands the intervention effects of dexamethasone on the expression and localization of AQP8 mRNA in oligohydramnios placenta and fetal membranes, and explores the targeted regulation with AQP. Therefore, it provides a new way and new strategy for the prevention and treatment of high-risk pregnancy caused by oligohydramnios.Methods:(1) Cells of placenta and fetal membrane were cultured and divided into study group and control group.(2) Extract total RNA of those cells after respective addition of different doses of dexamethasone (0μg/ml, 1μg/ml,5μg/ml,10μg/ml, and 50μg/ml) and incubation for 24 h.(3) Expression of AQP8 mRNA in placenta and detection of fetal membranes by RT-PCR.(4) The location of AQP8 in placenta and detection of fetal membranes by immunohi stochemi stry.Results:(1) Expression of AQP8 mRNA was detected in both normal human placenta and fetal membranes.(2) Different concentrations of dexamethasone had enhanced regulation effects on the expression of AQP8 mRNA in WISH cells. The expression of AQP8 mRNA in amniotic epithelial cells and chorionic cells after addition of dexamethasone were improved, and the differences were statistically significant (P<0.05). Compared with control group, the expression of AQP8 mRNA in syncytiotrophoblast with treatment of dexamethasone in study group was similar, and there were no significant differences (P>0.05).(3) AQP8 distribution was detected in cytoplasm and membrane of human amniotic epithelial cells, chorionic cells and placental syncytiotrophoblasts.Conclusions:(1) Expression of AQP8 is detected in cytoplasm and membrane of oligohydramnios amniotic epithelial cells, chorion cytotrophoblasts and placental syncytiotrophoblasts.(2) Dexamethasone has a significant enhancement on the expression levels of AQP8 mRNA.(3) Dexamethasone may increase the expression of AQP8 in human amniotic epithelial cells and placental syncytiotrophoblasts to make certain effect on the steady of amniotic fluid, mechanism needs further investigation.
