| ObjectiveThe development and progression of gastric cancer and its growth, invasion and metastasis is involved in the amplification and abnormal expression of multiple genes. Until now, there is not a reliable biological marker for determination of tumor malignancy and prognosis of patients, is not the ideal biological therapeutic target. HER-2/neu is one of the epidermal growth factor receptor (EGFR) family member. Studies have shown that in cancer with HER2/neu overexpression, and its poor prognosis and resistance to chemotherapeutic drugs play a role depends on the activation of PI3K/Akt signaling pathway, and Akt2 overexpression is closely related with invasion and metastasis. But there is no report about correlation of HER2/neu expression with Akt2.In this study, Akt2 and HER2/neu expression was detected in gastric cancer and control gastric tissues by immunohistochemical method, it was also analysed the effect and relationship of Akt2 and HER2/neu and its mechanism during gastric cancer development, and the result is helpful for cancer diagnosis and prognosis determination.Materials and methodsTo detect the expression of HER2/neu and Akt2 in 80 cases of gastric cancer and 40 control gastric tissues (chronic gastritis, gastric ulcer) by immunohisto- chemical method, and to analyse the relationship of them with Cancer clinical stage, differentiation degree, invasion and lymph node metastasis.Results1. HER2/neu expression in gastric cancer tissues The positive rate of HER-2/neu in gastric cancer tissues was 17.50%, the difference was statistically significant when compared with control group (P <0.05).2. Akt2 expression in gastric cancer tissues The positive rate of Akt2 expression in gastric cancer tissues is 67.50%, and that in control group is 5.00%, there is a statistically significant difference (P <0.01). 3,The relationship of HER2/neu and Akt2 expression with clinical pathological parametersThere is no relation between HER-2/neu protein in gastric cancer and clinical stage, the depth of invasion, lymph node metastasis, the differentiation degree of tubular adenocarcinoma (P> 0.05). Akt2 expression in gastric cancer tissues is related with clinical stage and lymph node metastasis, Lauren type, the differentiation degree of tubular adenocarcinoma (P <0.01), but not its WHO classification and invasion depth (P> 0.05). The expression of HER-2/neu in gastric cancer tissues is positively correlated with Akt2 expression (P <0.05).Conclusion1. HER-2/neu and Akt2 protein expression in gastric cancer tissues increases.2. Akt2 expression in gastric cancer tissues is related with the clinical stage of cancer, lymph node metastasis, Lauren type, the differentiation degree of tubular adenocarcinoma.3. The expression of HER-2/neu in gastric cancer tissues is positively correlated with Akt2 expression. |
