BackgroundPsoriasis is a kind of multigene genetic diseases which is affacted by genetic and environmental factors,this inflammatory erythematous scaly skin disease is involved of many inflammatory factors. Higher prevalence of psoriasis, many young adults infringement, easy to relapse, longer course, a great impact on physical and mental health patients.Currently, many treatments of psoriasis and methods have been used, but failed to prevent or to prevent recurrence by drugs.Retinoic acid drugs have been widely used in the clinical treatment,and on the good results,but it has poor targeting and specific side effects.Studies have demonstrated that peroxisome proliferator-activated receptorγ(peroxisome proliferator-actived receptorγ, PPARγ) in psoriatic skin lesions than normal tissues to low, and psoriasis patients PPARγexpression in non-lesion area compared with normal skin, no significant differences in expression.PPARγand Retinoic acid receptors,belong to the nuclear hormone receptor superfamily,which have anti-inflammatory and anti-proliferation fuctions in psoriasis and other diseases. And PPARγ's ligand such as troglitazone is also found the effective treatment of psoriasis in the recent years. In recent years, the etiology of psoriasis, pathogenesis and the therapeutic attention PPARγgradually become the hot topic.ObjectiveThis study was conducted by Acitretin capsule on moderate to severe chronic plaque psoriasis' patients before and after treatment,to found the expression changes of peroxisome proliferator-activated receptorγ(peroxisome proliferator-actived receptorγ) in the psoriatic lesions. Through this experiment, we want to explore the role of PPARγin the development of psoriasis, the mechanism of retinoic acid in the treatment of psoriasis, and the mechanism of side reaction of retinoic acid .Thus, we could research new drugs which can prevent and cure psoriasis.Methods The clinical and pathological diagnosis of 18 patients with psoriasis, in the application of Acitretin capsule around before and after surgery were drawn in the lesion, derived specimens preserved in 2 parts, part of the solution in 10% formalin fixed, paraffin pack to be buried in the methods used for immunohistochemical detection of lesions in psoriasis before and after treatment with PPARγprotein levels; another part of the refrigerator placed in -80°C cold to stand in the RT-PCR of mRNA expression of PPARγwas detected level.ResultsTissue lesion of psoriasis patients before and after treatment by immunohistochemical staining, positive cells mainly in the basal cell layer and the upper spinous layer cells, mainly for nuclear color, brownish yellow. And in the application of psoriasis patients before treatment of Acitretin capsules,the expression of PPARγwas significantly lower than after administration of the PPARγexpression intensity (χ2 = 9.837, P <0.01); PPARγin the RT-PCR, semi-quantitative analysis showed that the expression of PPARγmRNA before treatment was significantly lower than the skin after treatment tissues (t = 26.13, P <0.001).ConclusionAfter the treatment of Acitretin capsule,the expression of PPARγin the quantity of tips and the changes of location in the psoriasis skin lesions ,it is found that the retinoic acid can increase the quantity of PPARγ, and PPARγmay play a role in the development of psoriasis .Also,PPARγor its ligands may be the targets which can block the inflammatory of psoriasis.
