Objective:By observing the diarrhea-predominant irritable bowel syndrome (referred to as IBS) rat model of plasma and colon tissue of vasoactive intestinal peptide (referred to as VIP) expression levels change and the cecal mucosa mast cells (MC) count changes, to explore traditional Chinese medicine-r-elated irritable bowel capsule mechanism.Method:1. making model:randomly selected 60 male SD rats weighing (160±20) g. Application of restraint-Brake stress method and chemical stimulation of the Combination Prepar-ation of colonic enema diarrhea-predominant irritable bowel syndrome model. Modeling process is complete, the rats of model group were to assess the sensitivity of visceral nerves (abdominal retraction reflex) to assess the success of our mo-deling.2. Animal grouping and drug delivery:successful model after the rats were randomly divided into 6 groups (normal control group, model control group, western medicine, Chinese medi-cine low dose treatment group, traditional Chinese medicine in the treatment groups, high dose group of Chinese medinc- ine.) Equal volume of each group are 10ml/kg gavage. One normal control group and model control group were fed with saline, western medicine group with 1.8mg/ml of concentrati-on may Schutt liquid gavage, capsule medicine bowel with low-dose treatment group the concentration of 0.225g/ml bo-wel capsule liquid gavage, capsule dose of Chinese medicine treatment of irritable bowel group with the concentration of the irritable bowel capsule 0.45g/ml liquid gavage, Chinese medicine treatment of irritable bowel high dose group with t-he concentration of the irritable bowel 0.9g/ml capsule liquid gavage. The group was given both day 1 for 2 weeks.3. Index test:①rectum were observed in rats when they belly balloon retraction reflex threshold and when the capacity exp-ansion within 3min when the volume of 1.0ml of abdominal c-ontractions.②by enzyme-linked immunosorbent assay (Elisa) detected in plasma and colon mucosa, VIP expression.③by toluidine blue staining of their cecal mucosa MC staining, light microscope, counting 10 high power field in the MC number.Results:1. Model group, low dose group of Chinese medicine, Chinese medicine treatment in rats caused dose abdominal retraction reflex threshold was significantly lower than the capacity of the normal control group, significant difference (P<0.05), while the western medicine group Chinese high-dose treatment group and control group, no significant difference (P> 0.05); Each treatment group compared with the control group, significant difference (P<0.05); the treatment group and western medicine treatment group, no significant difference (P> 0.05). Model control group, low dose group of Chinese medicine, Chinese medicine in the treatment groups compared with the control group,1.0ml of water injection within 3min when the number of rats was significantly higher abdominal reflex, significant difference (P<0.05), while the other treatment groups compared with the control group, no significant difference (P> 0.05);The treatment group compared with the control group, significant difference (P<0.05); the treatment group and western medicine group, no significant difference (P> 0.05). The results showed that the capsule on Intestinal Irritable Bowel sensitized behavioral changes caused by the resumption of role play.2. Model control group, western medicine, Chinese medicine low dose treatment group, traditional Chinese medicine in the treatment groups compared with the control group, the content of VIP in rat plasma were significantly different (P<0.05), while the high dose group and traditional Chinese medicine the normal control group, the content of VIP in rat plasma was no significant difference (P> 0.05); medicine in the treatment group compared with the control group, the content of VIP in rat plasma were significantly different (P<0.05); the traditional Chinese medicine western medicine group and the group was, in rat plasma VIP levels were significantly different (P<0.05). The results show that the model group, plasma VIP levels were higher than normal control group, easily excited capsule can significantly reduce the model group, plasma VIP levels.3. Model control group, western medicine, the TCM treatment group compared with the control group, rat colon, VIP levels were significantly different (P<0.05); traditional Chinese medicine in the treatment group and model group, rat colon tissue content of VIP significantly different (P<0.05); the treatment group and western medicine treatment group was compared with colon tissue VIP content was significantly (P <0.05). The results show that the model group, VIP content in the colon of the control group were higher than normal, easily excited capsule can significantly reduce the model group, VIP content in the colon.4. Model control group, western medicine, Chinese medicine low dose treatment group, traditional Chinese medicine in the treatment groups compared with the control group, rats in the cecal mucosa MC counts were significantly different (P<0.05), while the high dose of Chinese medicine group and control group, rats in the cecal mucosa MC count was no significant difference (P> 0.05); western medicine, Chinese medicine in the treatment groups, Chinese high-dose treatment group compared with the control group, rat cecal mucosa tissue MC counts were significantly different (P<0.05). The results show that the model group in the colonic mucosa MC counts were higher than normal control group, easily excited and Western have Schutte capsules were able to model rats significantly decreased in the colonic mucosa MC count.Conclusions:1. Diarrhea-predominant irritable bowel syndrome model rats VIP in plasma and colon tissue were significantly higher, indicating that vasoactive intestinal peptide (VIP) may be the cause of irritable bowel syndrome, an important biologically active substances.2. Diarrhea-predominant irritable bowel syndrome model rats, cecal mucosa was significantly higher in MC counts, indicating that irritable bowel syndrome may occur with the increase of MC associated with activity3. Bowel capsule significantly improved compared with diarrhea-predominant irritable bowel syndrome model rats the capacity of abdominal retraction reflex threshold and reduce the unit of time equal volume of rat abdominal rectal balloon times of reflection. Show that the capsules can reduce diarrhea irritable bowel irritable bowel syndrome model rats visceral nerve hypersensitivity.4. Bowel capsule can reduce diarrhea-predominant irritable bowel syndrome model rats plasma and colon tissue VIP content. That may be one of the therapeutic mechanism and by lowering plasma VIP levels in the colon to reduce the sensitivity of visceral nerves, which reduces the sensitivity of intestinal external stimuli.5. Bowel capsule can reduce diarrhea-predominant irritable bowel syndrome model in the rat colonic mucosa MC count. Another treatment that its mechanism may be to reduce infiltration and inhibition of MC MC activities, reduce the associated release of biological material in order to achieve the purpose of ease abdominal pain and diarrhea.6. Bowel capsule of Chinese medicine in improving the therapeutic effect of diarrhea-predominant irritable bowel syndrome symptoms more prominent role. And its treatment within a certain range is proportional to its dose and high dose was more significant.
