Objective: Through observe oxidatie stress and NF -κB c - Rel/RelA ratio′s change under vitamin K2 , to explore the mechanisms of apoptosis in human prostate cancer PC-3 cells induced by Vitamin K2.Methods: PC-3 cell was treated with different concentration VitK2(0,10,20,40and80μmol/L).And then ,the proliferation of cells were detected by cck-8;the apoptosis of PC-3 cells were detected by PI-AnnexinV double staining assay;GSH containing was detected by Glutathione Reduced Assay Kit;the expressions of RelA and c-Rel were determined by western blotting.Results: With the increase of concentration , PC-3 cell growth inhibition rate and apoptosis rate increased gradually ;Microplate analysis results showed reduced glutathione levels with Vitamin K2 concentration increases;with the increase of Vitamin K2 concentration ,RelA gene expression of antiapoptotic raised after the first cut and c-Rel was obviously up-regulated,c-Rel/RelA ratio gradually increasing.Conclusion: Vitamin K2 may be started NF-κB by oxidative stress to adjust RelA and c-Rel expression to induce PC-3 cell apoptosis.
