Objective: to investigate the protection of Gas6 from H9c2 cells apoptosis in ischemic preconditioningMethods: The H9C2 cells were randomly divided into five groups: normal control group ; ischemia-reperfusion group; ischemic pre-conditioning(IPC)group; Gas6 group and Gas6+LY294002 group. serum LDH was detected with kit. Cell apoptotic rate was evaluated by flow cytometry (FCM). The Gas6 mRNA was examined by the real-time PCR and P-Akt and P-FoxO3a protein expressions were examined by western blot.Results: compared with IR group, Gas6 mRNA expression in IPC group significantly increased (P〈0.05), serum LDH level and the cell apoptotic rate (P〈0.05)significantly decreased in IPC group and Gas6 group, meanwhile P-Akt and P-Foxo3a protein expressions significantly increased(P〈0.05); compared with Gas6 group, serum LDH level , the cell apoptotic rate and P-Akt and P-Foxo3a protein expressions were not different in IPC group(P〈0.05),but serum LDH level and the cell apoptotic rate significantly increased in Gas6+LY294002 group,meanwhile P-Akt and P-FoxO3a protein expressions significantly decreased(P〈0.05).Conclusion: Gas6 had a Cardioprotection in ischemia preconditioning. This may be mediated by a Phosphatidylinositol3-Kinase/Akt/FoxO3a Survival Pathway.
