Objective To compare the anti-cicatrization effect of interferonα-2b (IFNα-2b) with 5-Fluorouracil (5-Fu) in glaucoma filtering surgery.Methods 52 patients (56 eyes) with glaucoma were randomly studied by treatment with IFNα-2b(28 eyes) or 5-Fu (28 eyes)following trabeculectom ,4 were bilateral, the former was designed to use IFNα-2b, and the latter apply 5-Fu .and they all received the surgery the first time.The IFNα-2b treated eyes and The 5-Fu receiving eyes were intraoperatively administered respectively with 5×105IU and 12.5mg/ml via a sponge under the conjunctival flap for 4 minutes. And acupuncture separation combined with injection of IFNα-2b or 5-Fu around the filtering belb after glaucoma filtering surgery, Observe the intraocular pressure (IOP),visual acuity,complications etc.The follow-up was 4 months.Results Comparing the postoperative with preoperative,the intraocular pressure(IOP) of the two groups significantly declined (P<0.01). The decrease of visual acuity was seen in 39.3% in 5-Fu group,whereas only 10.7% in IFNα-2b group. The difference between the two groups was remarkable (χ2=6.095,P<0.05).The eyes with IFNα-2b tended to form type II blebs were according to Kronfeld classifi- cation, whereas type I blebs were commonly seen in 5-Fu treated eyes (χ2=8.096,P<0.05). the degree of corneal eraosion in IFNα-2b was significantly lower than that in 5-Fu group (P<0.05) . Conclusions Application IFNα-2b and 5-Fu in glaucoma filtering surgey all can prevent the bleb from scar formation,reduce the risk of failure of glaucoma filtering surgery. and the complications of IFNα-2b were more rare than 5-Fu, It is the idea method for treatment of glaucoma.
