Antitumor Effects Of Saponin Extract From Patrinia Villosa (Thunb.) Juss On Mice Bearing U14 Cervical Cancer

Natural anti-cancer drugs gradually become the focus of research,because of its toxic side effects,biological activity and non-resistance, etc.Patrinia (Herba Patriniae) as Valerianaceae Plant Patrinia (Patrinia villosa Juss.) of the whole plant, widely used in clinical therapy,such as early acute appendicitis, unknown throat, cervical erosion and cancer. However,studies on the Patrinia saponins (Saponins of Patrinia villosa (Thunb.) Juss PVJ-S) against cervical cancer has not been reported ever.In this experiment,by establishing solid tumor model of U14 cervical cancer,detection of tumor growth and immune-related biochemical indexes,the results as .followings.1.By different concentrations(12.5 mg/mL,25 mg/mL and 50 mg/mL) of Patrinia saponin extract affect on Hela cells were obtained inhibition rate were 67.7 %,69.8 % and 70.1 % through MTT test.The results show that Patrinia saponin extract can inhibit the proliferation of Hela cells. Hoechst staining results of Hela cells compared with the control group,the Patrinia saponin extract of Hela cells caused more morphological changes of apoptosis, revealed Patrinia saponin extract may inhibit the proliferation of Hela cells through the induction of apoptosis.2.Through the establishment of solid tumor model of U14 cervical cancer tumor-bearing mice,treated with different concentrations (low dose 250 mg / kg and high dose 1 000 mg/kg) Patrinia saponin extracts were orally treated, respectively, get the inhibiting tumor rate was 54.6 % and 42.1 %, compared with the control group shows significant difference (p<0.05),and the index of immune organs can significantly increased(p<0.05).3.Research of solid tumor model of U14 cervical cancer,and normal saline group as negative control, cyclophosphamide (CTX) as a positive control, continuous processing 15 d, Patrinia saponin extracts can resistance in mice Oxidative capacity and aspects of immune regulation of the possible anti-tumor mechanism .The results show that Patrinia saponins high dose and low dose could significantly increased SOD activity and CAT activity,compared with the negative control group(P <0.01),which revealed Patrinia Saponins can be improved Oxidation state. 4.Through the three detection kits(Annexin V-FITC apoptosis detection kit, Hoechst33342 apoptosis and DAPI fluorescence staining kit) for tumor cells,research shows that Patrinia saponin extract promoted the apoptosis of tumor, clearly observed the generation of apoptotic bodies, which can reveal Patrinia saponin extract can be related to apoptosis induced by inhibition of tumor growth.The study was approved by AB-8 macroporous resin purified Patrinia saponin extract,through the establishment of solid tumor model in mice U14 cervical cancer,and analysis of tumor growth and its associated immune biochemical indicators, to reveal Patrinia saponins anti-cancer mechanism to provide a new basis for further study of natural anti-cancer drugs.
