| Object and method:A number of 171 cases were randomly selected as normal glucose tolerance (NGT), of which 86 cases were from Han and 85 cases were from Korean. Likewise, a number of 225 cases were randomly selected from type 2 diabetes-mellitus (T2DM), of which 131cases were Han and 94 cases were Korean. According to the carotid artery intima-media thickness (IMT), the cases of T2DM were divided into four groups:(1) Han with carotid atherosclerosis of which IMT is larger than 1 mm or with Plaque; (2) Han with carotid atherosclerosis of which IMT is less than 1 mm or without Plaque; (3) Korean with carotid atherosclerosis of which IMT is larger than 1 mm or with Plaque; (4) Korean with carotid atherosclerosis of which IMT is less than 1 mm or without Plaque. Both group of Han and Korean were divided into two groups, according to the different genotype of GG and GT+TT. Carotid artery IMT were determined by Color Doppler and posteriority mtDNA A3243 G mutation rates were analyzed by TaqMan probe Real Time PCR. The polymorphism of genotype of adiponectin SNP+276G/T were tested by PCR-Taqman. The results were statistically analyzed by SPSS 17.0 software.Result:1. In Yanbian the acquired mitochondrial DNA 3243A/G somatic cell mutants of the NGT group (0.046±0.449V.S0.739±0.738) is different from that of T2DM (P<0.05);likewise the acquired mitochondrial DNA 3243A/G somatic cell mutants of the IMT group(0.279±0.255V.S0.938±0.565) is significantly different from the group of no IMT (P<0.05).2. The genotype of adiponectin SNP+276GG of with T2DM the acquired mitochondrial DNA 3243A/G (0.596±0.650V.S 1.061±0.532) is different from the genotype of adiponectin SNP+276GT/TT (P<0.05); also there is significant different between the genotype of adiponectin SPN+276GG of the T2DM with IMT (0.632±0.603 V.S0.964±0.342) and the genotype of adiponectin of SPN+276GT/TT of the T2DM with IMT (P<0.05). This results of polymorphism of genotype of adiponectin SPN+276GG and SPN+276GT/TT, shows that there is no difference between Han and Korean on the acquired mitochondrial DNA 3243A/G of the T2DM with IMT. Conclusion:1. In Yanbian the chronic oxidative stress level of the T2DM is higher than the NGT group.2. The long-term oxidative stress level of the T2DM with IMT is higher than the T2DM.3. There is association between the genotype of adiponectin SNP+276G/T and the long-term oxidative stress in both of T2DM group and the IMT group, however, no difference between Han and Korean in Yanbian area was observed. |
