Objective:To obtain the simulative aim oligopeptide which has the better homology to Exendin-4 using the sift technology from peptide library. Synthetize, purify and identify the aim oligopeptide. Demonstrate the hypoglycemic effect of the simulative oligopeptide by proliferation experiments in vitro and hypoglycemic experiments in vivo.Methods:1. Obtain the simulative aim oligopeptide named 12PA after four times sifting from the peptide library of phages. Synthesize,purify and identify 12PA.2. Test the every well of 96 well plate where the RIMm-5F cells were mixed with the sample of 12PA.3. Multiple STZ injection of small dose to the Wistar rats which were divided into several groups at random.Inject the rats peritoneally once a day according to the blood glucose levels of every group. Five weeks later,kill the rats and detect the relative indexes.Results:1. Sift and synthesize the simulative aim oligopeptide.Certify the correctness of the compound above-mentioned by HPLC and MS.2. Demonstrate the synthesized oligopeptide having the role of proliferation to the RIMm-5F cells in cell experiments.3. Demonstrate the synthesized oligopeptide having the hypoglycemic effect to the rats with typeâ…¡diabetes by the experiments in rats and the glucose tolerance experiments.Conclusions:Demonstrate the simulative oligopeptide named 12PA having a certain hypoglycemic effect to typeâ…¡diabetes as Exendin-4 which has the high homology to 12PA. The next plan is to better the short half-life time and the instability of 12PA.
