Type 2 Diabetic Rats Hyperhomocysteinemia The Intracavernous H <sub> 2 </ Sub> S / Cse System Impact

Objective: In order to study the relation between homocystine (Hcy) in type 2 diabetic rat and H2S/CSE system in penile corpus cavernosum ,with an attempt to explore the mechanism of diabetes mellitus induced erectile dysfunction.,Methods: Ten rats were randomly selected. From fifty Wistar masculine rats of 3 months supplied by normal feeding. as control group(A. group; n=10). Other rats were fed by high-fat and high-glucose. After 4 weeks streptozotocin (30mg/Kg) were injected into caudal vein. Selected thirty diabetic rats, divided into three groups: diabetic rats group (B group; n=10). ; Diabetic rats treatment with insulin group (C group; n=10). ; diabetic rats treatment with folic acid group ( D group; n= 10) ; A group supplied by normal feeding ; B,C,D groups were fed by high-fat and high-glucose . Injected with apomorphine on the 12th week, erections of the rat were observed and noted .The levels of the total plasma homocystine were measured by method of cycle enzyme. The rats were sacrificed to take the penile corpus cavernosum, the penile corpus cavernosum of rats in each group were randomly devided to two group, one group were used to measure the CSE and the other group were used to measure H2S in the penile corpus cavernosum.The cystathionineγ-lyase (CSE) were observed under laser scanning confocal microscope.The Hydrogen sulfide (H2S) were measured by the remnant penile corpus cavernosum.Results: The HCY in Type 2 diabetic rats was significantly increased , but the number of penile erection, cystathionineγlyase (CSE) and the H2S in the rat corpus cavernosum was decreased, compared with the normal group was statistically significant (P < 0.01). Insulin and folic acid therapy group the CSE and the H2S content in penile cavernous tissue comparison with non-treated group was significantly higher, significantly increased the number of penile erection, was statistically significant (P <0. 01).Conclusions: Type 2 diabetic rats with high plasma HCY levels have a negative correlation with the CSE and H2S in the penis , this may be the pathogenesis of type 2 diabetic rats erectile dysfunction. Diabetic insulin and folic acid can help to improve erectile dysfunction.
