Objective:To observe replicative senescence of cells extracted from the endplate in the intervertebral discs and Explore cartilage endplate in the pathogenesis of degenerative process。Methods:The intervertebral disc endplate were obtained from 20 patients with disc herniations; from 25 patients undergoing posterior surgery for either back pain due to degenerative disc disease (n=20) or spondylolisthesis (n=5) and from five patients with vertebral body fracture。Tissue sections were assessed for the degree of cell senescence (using the marker senescence-associated-β-galactosidase (SA-β-Gal)) and the number of cells present in clusters, But hip Osteoarthritic human articular cartilage was examined as a positive control from four patients。Results:There were significantly more SA-β-Gal positive cells in herniated discs (14.0±11.78%of cells) than those with degenerative disc disease spondylolisthesis and vertebral body fracture(3.03±3.51%, P<0.0001). In herniated disc of cells in clusters (defined as groups of three or more cells) (19.8±13.68%) were more frequently SA-β-Gal positive than single or double cells (8.20±5.61%, P<0.02)。Conclusion:The present study indicate degeneratie lumbar cartilage endplate cells are subject to senescence, especially in herniated disc of discs endplate cells.And replicative senescence may be involved in the aetiopathogenesis of age-related intervertebral disc degeneration。...
