Purpose: Research the saponins from fruit of Tribulus terrestris L metabolized by human intestinal bacteria and rat, analyze variations between prototype saponins and metabolites, separate and identify metabolites by human Intestinal bacteria. Explore the relativity of structures between prototype saponins and metabolic products. To elucidate metabolic processes in vivo and the effactive ingredients that really laid the foundation.Methods: Respectively by means of in vitro culture and blood analysis method is used to study metabolites of the saponins of fruit of Tribulus terrestris L, through TLC, HPLC–ELSD, HPLC-APCI-MS analysis method,determine the metabolites of the saponins. Enriched cultivation, separate metabolites by silica gel column and identity them by TLC, HPLC-ELSD etc., and further by chemical methods and modern spectroscopic techniques (1~H NMR, (13)~C NMR, APCI-MS, HPLC-MS, etc.) to identify the structure of metabolites.Results: The total saponins from fruit of Tribulus terrestris L is decomposed easily,and nine metabolites are found as hecogenin, manogenin, hecogenone, 25R-spirostan-3, 5-diene-12-one, ruscogenin, gitogenin, diosgenin, chlorogenin, 25R-spirostan-3,5-diene.From the blood of rats that are feed with the saponins from fruit of Tribulus terrestris L, four genins are determined as 25R-spirostan-3,5-diene-12-one, hecogenin, hecogenone, 25R- spirostan-4-ene-2,3-dihydroxy-12-one.Five compounds are isolated from the metabolites, the spectral structures are identified as: nonadecanoic acid (Ⅰ), tigogenin (Ⅱ), hecogenin (Ⅲ), gitogenin (Ⅳ), manogenin (Ⅴ).Conclusion: This study shows that saponins of Tribulus terrestris fruit can be metabolismed by intestinal flora easily, compenents of high polarity saponions metabolismed to compenents of low polarity aglycones. It is accorded with the rule of natural glycosides meabolismed by intestinal flora. During the test of the blood sample, the chromatograph shows a new peak, which is unknown. We need to do further reseach to identification.
