Objective:We carried on systematic toxicity researches of Arisaema by conventional toxicological studies based on the current understanding of the toxicity of Arisaematis Rhizoma. The toxic characteristics and mode of action were clarified, which can guarantee reasonable processing of Arisaema and provide experimental basis for the clinical uses of drugs.Methods:(1)The extraction process of Arisaema'were optimized by orthogonal design method; (2)LD50 of extract of Arisaema in oral doses were determined by acute toxicity test; (3)Systematic studies were carried on the general toxicity of extract of Arisaema by long-term toxicity test.Results:(1)Through L9(34)orthogonal experimental design, the yield of alcohol extracts and total flavonoid contents were chose as the index, and the main factors in the extraction, such as ethanol concentration, solvent volume and reflux time, were investigated. The optimum extraction conditions were:8 times the amount of 60% ethanol, reflux extraction twice, each time 30min, and above process were stable;(2)An oral administration to mice, LD50 was measured 155.78g/kg (crude drug).(3) Continuous administration of the extraction of Arisaema raw materials for 30 days to rats, blood regular indexes and biochemical indicators in drug group were compared to control group, and some indicators showed significantly differences; Histopathological slice exhibited no significant lesions of other organs except the significant increase of intercellular spaces of cardiocytes and obvious striction of alveoli.Conclusion:Ethanol extract of Arisaema in the administration of oral dose existed obvious symptoms of acute toxicity, the toxicity mainly concentrated in heart,lung and kidney. |
