Objective"Ju-Ju", the dried leaves and stems of Cichorium intybus L.(C. intybus), which belongs to the family Compositae, is a Uyghur medicine commonly used in Xinjiang of china. C. intybus is produced in Europe and Asia. In Europe, it is used as vegetable, substitute of coffee and medicinal herb against liver damage. The seed of C. intybus is used asemmenagogue in Indian. Chinese use herb of C. intybus to protect liver and strengthen the stomach in Inner Mongolia and Xinjiang.Previous chemical investigations have indicated the presence of sesquiterpene, steroid, coumarin, and carbohydrate in the root of C. intybus. Pharmacological investigation demonstrated that it has the effects of hepatoprotection, lowering blood lipid and encourage digestion. In order to identify active site against hepatic damage, the present study was to investigate the compound of C. intybus and the effects on liver function.Animal and human body are facing a very common and severe damage that is the liver oxidative damage that cause animal and human body apolexis and other diseases, aggravate organism apolexis and affection. At present, it has more anti-liveroxidativedamaged medicine, but most are obtained difficulty, low purity, unidentify effective constituent, and higher price. So it is very urgent and significance that developing an anti-liveroxidativedamaged medicine that is wide source, high purity, constituent identify, good effect, safety, asepsis.Methods1. Induction of animal Model:Adopt CCl4 to induce hepatic fibrosis, use C. intybus to interfere in the rats of hepatic fibrosis by CCl4 induced. Use the method of biochemistry and pathematology to observe the protection of C. intybus for hepatic fibrosis induced by CCl42. Animal groups and administration:60 rats were randomly divided into 5 groups, namely the model group, Fufangbiejiarangan film positive control group, low dose group of C. intybus, middle dose group of C. intybus, high chicory dose group of C. intybus. Model group was injected 1mL/kg 40% CCl4- oil from the fifth week,2 times per week. Other groups were injected in the abdominal cavity 1mL/kg 40% CCl4- oil (2 times per week), while oral administration,1 time per day for 4 weeks. Doses were:solvent control group was given 10mL/kg saline, positive control group 750 mg/kg, low dose 2.0g drug/kg, the dose of 6.0g drug/kg, high dose group 18.0g drug/kg.3. Sample collections:The end of the eighth week of experiment, after administration in fasting the last 20h, blood behind the eyes off the neck were killed, serum -20℃preservation. Right lobe of the liver which was fixed in 10% formaldehyde solution for pathological examination (HE staining, Masson trichrome staining), and the remaining liver stored in liquid nitrogen, the standby4. Detection of indexes:Aspartate aminotransferase (AST), alanine aminotransferase (ALT), hyaluronic acid (HA), laminin (LN), hydroxyproline (HyP), reduced glutathione (GSH), superoxide dismutase (SOD) and malondialdehyde (MDA)Results1. C. intybus can significantly inhibit serum ALT, AST, fatty degeneration of liver cells and reduce the infiltration of inflammatory cells, that indicated it has the protection effect of the liver cells.2. C. intybus can inhibit HA, LN, HyP higher level, and pathological observations suggest that it can inhibit the proliferation of fiber, indicating that it can really inhibit the synthesis of collagen fibers in the liver.3. C. intybus also significantly reduced liver fibrosis abnormal increase in liver tissue MDA, increased SOD and GSH content. That suggests its effect of inhibit hepatic fibrosis may be correlated with inhibit production of MDA and free redical scavenger.ConclusionThe present study was a profound reseach about the constituents from C. intybus. and protective effects against liver injury, its methods were reliability, precise accuracy and easy operation. In thesis, there is lots of information about pharmacodynamics, pharmacology and clinical research of the plant, which has estab-lished solid foundation of the plant to further its development and reseach.
