| ObjectiveTo study the expression of FAK (focal adhesion kinase), P-FAK(Tyr397) and PTEN(phophatase and tensin homolog deleted in Chromosome 10) in gastric adenocarcinoma (GA) and their relationship with clinical pathological characteristics and to investigate their roles in carcinogenesis, development, invasion and metastasis ofGA.MethodsThe expression of FAK, P-FAK(Tyr397) and PTEN in 100 cases of GA tissue and 30 cases of adjacent normal gastric tissue was detected by immunohistochemistry.Results(1) Differences were observed in the the positive expression rates of FAK, P-FAK(Tyr397) and PTEN protein between GA (71%,62%,53%) and adjacent normal tissue (43%,20%,97%) (P<0.05).(2) In GA, the expression of FAK and P-FAK(Tyr397) was positive correlated with depth of invasion, lymph node metastasis and TNM stage(.P<0.05), negative correlated to the degree of differentiation (P<0.05). While the expression of PTEN was negatively correlated with depth of invasion-lymph node metastasis and TNM stage(P<0.05), positively correlated to the degree of differentiation (P<0.05). The expression level of them is independent of age and sex(P>0.05).(3) By using Spearman rank correlation analysis, the expression of PTEN was negatively correlated with FAK (r=-0.457, P<0.05) and P-FAK(Tyr397) (r=-0.497; P<0.05) expression in GA, but the FAK expression was positively correlated with the P-FAK(Tyr397) expression (r=0.536; P<0.05).Conclusions(1) The high level expression of FAK, P-FAK(Tyr397) and the low expression of PTEN were positively correlated with carcinogenesis, development, invasion and metastasis of GA.(2)There existed negative correlation between the expression of PTEN and FAK,PTEN and P-FAK(Tyr397) in GA, suggesting they might be mutual checks during the development of GA.(3) The phosphorylation level of Tyr397 may reflect the activation of FAK protein in cells, combination of FAK and P-FAK (Tyr397) are in favor of prediction the genesis, development and prognosis of tumor.(4) The combined detection of FAK, P-FAK (Tyr397) and PTEN protein has positive effects on predicting the diagnosis, malignant degree and prognosis of GA... |
