The cell abnormality proliferation induced by apoptosis mechanism disorder is one of the mechanisms of breast cancer development. p53 upregulated modulator of apoptosis found in 2001 is a significant promoter gene of apoptosis.It has confinned in vitro that,PUMA gene can be induced by p53 or DNA injury medicine and radioactive ray,and then initiated cell apoptosis.The expression product of PUMA gene is an Bcl-2 family and BH3-only subfamily member,through a series of cytokine signal transmission, at last change the permeability of mitochondrial,then trigger cell apoptosis by mitochondrial pathway. PUMA is one of the hot pot of tumor therapy and tumorigenesis mechanisms in latest years.But there is no reports about the expression of PUMA in breast cancer.Objective To study the expression of PUMA mRNA in breast neoplasms tissue, the relation between PUMA mRNA and the clinic pathological characteristics of breast cancer, and its relation to Bcl-2 and Bax.Methods The tumor tissue from 30 patients declared breast neoplasms without chemotherapy or radiotherapy before operation are assigned as group A; the group B contains the same 30 patients in Group A, but the tissue are harvested in the operation after undertook FEC chemotherapy; the tissue from a total of 20 patients with hyperplasia of mammary glands are assigned as group C; and the expression of PUMA mRNA is observed with RT-PCR technique, Bcl-2 and Bax, immunohistochemical method. The clinic pathological characteristics include age of the patients; size, histopathology classification, TNM stages and lymphnode metastasis of the tumor are statisticed.Results There is no significant difference of PUMA mRNA expression level in breast neoplasms(group A) and in benign breast lesions (group C), and the expression is up regulated in the patient undertook FEC chemotherapy(Group B). There is significant relationship between PUMA mRNA expression and lymphnode metastasis, no significant relationship with clinic pathological characteristics, such as age of the patients, size, histopathology classification, and TNM stages of the tumor. The expression of PUMA mRNA is not related to the expression of Bcl-2 and Bax.Conclusion Breast neoplasms is not induced by the Low expression of PUMA. FEC chemotherapy may up regulate the expression of PUMA mRNA, and then induce the apoptosis of tumor cell. This may be the mechanism of it's therapeutically effect. The low expressions of PUMA lead to invasiveness increasing and metastasis. PUMA mRNA might regulate the functional condition of Bcl-2 and Bax, not the quantity of them. |
