Objective:To observe the effects of LEF on MCP-1 and IL-1βin endometriosis rats and therapeutic effect on experimental endometriosis and to approach its probable mechanism.Method:1.60 sexually mature females are not mating, Sprague-Dawley rats (180-200g) were used for the experiment.45 of them were selected randomly, by transplanting with autologous endometrium, endometriosis was surgically induced in them.2. After 3 weeks, collecting blood through caudal vein, taking suction peritoneal fluid, the rats underwent a second laparotomy, at the same time, the volume 1 of ectopic endometrium was measured.15 rats that did not attend the establishing model were acted as the blank control group, group C.36 rats confirmed EMT model successfully were randomly divided into 2 groups:group A, LEF treatment group, (n=18) and group B, model control group (n=18). After that, LEF was given to LEF group at concentration of 35mg-kg-1 by gavage per day; 0.9% saline was given to the model control group and blank control group, group C(n=15).9 rats that are established model of endometriosis unsuccessfully were foresaked. The treatment period is 3 weeks, and the change of diet,defecation,weight of the rats were observed.3. After treatment 3 weeks, collecting blood through caudal vein, taking suction peritoneal fluid, the rats underwent a second laparotomy, The volume 2 of ectopic endometrium was measured, and the ectopic endometrial tissue of LEF treatment group and control group were collected. The volume 1 and volume 2 of ectopic endometrium have a comparison in the LEF treatment group and model control group, The expressions of MCP-1 and IL-1βprotein in Ectopic endometrial tissue were evaluated by immunohistochemistry. The levels of IL-1(3 in the blood serum and peritoneal fluid were detected by radioimmunoassay (R1A);The weight of rats have a comparison among 3 groups.Results:1. The model of EMT in rats was established successfully,80%(36/45) rats were induced endometriosis.2. LEF could control the development of ectopic endometrium. Compared with the and model control group, the ectopic endometrium's volume of the LEF treatment group were obiously decreased (P<0.05);3. After treatment, the expressions of MCP-1 and IL-1βprotein in ectopic endometrium of LEF treatment group are both lowere than experiment control group (P<0.05);4. Before drug, the IL-1βlevel of blood serum of B group and C group had statistically significant difference (P<0.05);after drug, the level of IL-1βin blood serum of B group and A group had statistically significant difference (P<0.05).A group between before drug and after drug, the level of IL-1βin blood scrum had statistically significant difference (P<0.05).5. Before drug, the level of IL-1βperitoneal fluid of B group and C group had statistically significant difference (P<0.05);after drug, the level of IL-1βin peritoneal fluid of B group and A group had statistically significant difference(P< 0.05). A group between before drug and after drug, the level of IL-1βperitoneal fluid had statistically significant difference (P<0.05).6. During 3 weeks, the diet of the LEF treatment group rats is smaller than other groups from then on 10 th day, and several rats have a Diarrhea on 15th day, but the body weight were no significantly different among 3 groups(P>0.05);Conclusions:1. LEF could decrease the level of Interleukin-lbeta (IL-1β) and MCP-1 in EMT rats and could improve the microenviroment and control the development of ectopic endometrium. LEF could control the development of ectopic endometrium, and decrease the development of ectopic endometrium, which maybe the possible therapeutic mechanism.2. LEF maybe has relatively few side effects, which maybe have a relation to the dosage and quantity of taking it. It may be a potential therapeutic method for the treatment of endometriosis.
