Objective:In the procedure of extracting DNA from sacra conjunctive of the patients with chronic conjunctivitis and PCR, discuss clinical significance of polymerase chain reaction in the diagnosis of chronic conjunctivitis.Methods:We selected 36 patients (36 eyes) with chronic infectious conjunctivitis,24 patients (24 eyes) with chronic immunologic conjunctivitis, normal 22 people (22 eyes), take the serious side from patients with both eyes with conjunctivitis. Dividing DNA extracted from conjunctival sac secretion into two parts, adds bacteria primer and adenovirus primer for PCR amplification separately, observe the result in gel imaging analysis system after gel electrophoresis. At the same time observe the result of conjunctival sac germ culture of the first group.Results:1. Groupl (patients with Chronic infectious conjunctivitis) has 14 eyes amplified 520bp stripe, positive rate 38.89%;4 eyes amplified 1004bp stripe, positive rate 11.11%; 2 eye whose stripe possess, they are Statistically significant differences under Comparison (χ2= 7.40, P< 0.05).2. Group2 (patients with Chronic immune conjunctivitis) has 3 eyes amplified 520bp stripe, positive rate 12.5%; 1 eye amplified 1004bp stripe, positive rate 4.13%;1 eye whose stripe possess 520bp and 1004bp, positive rate 4.13%.3. Groupl (patients with Chronic infectious conjunctivitis) which also procedure bacteria culture method has 19 positive eyes, positive rate 52.78%and this method has little difference from PCR. (χ2=3.2, P>0.05).Conclusion:1. Bacterial infection is more than viral infection in chronic infectious conjunctivitis.2. Part of the Patients with chronic immune conjunctivitis may be allergic to the ingredients or product of virus and bacterium, or the a detected bacterium is the normal flora exists within the conjunctival sac.3. Although PCR test positive rate more bacteria culture method, but no obvious difference is not high demand for samples, and time is short, and economy of chronic conjunctivitis diagnosis has good application prospect, is a kind of alternative methods. |
