| Purposes: Long-term clinical practice, found that treat the sedative-hypnotic patients by application of PMC, it has a significant effect. As PMC is less reported, in order to search for the wu teng's effective part of the sedative and hypnotic , reduce the amount of medication,we carry the optimum extraction and purity technology study, combined total anthraquinones flavonoids of pharmacokinetic and clinical application,and make the purifications into dripping pill by solid dispersion technology.Methods: Using pharmacodynamic test,in the number of mice locomotor activity as the investigation index,to determine the sedative-hypnotic effect part of PMC by using L9 (34) orthogonal experiment. HPLC and ultraviolet spectrophotometry was used to analyze the content of the total anthraquinones flavonoids, to optimize the extraction technology of the sedative and hypnotic effective.And purify the crude extract of PMC by macroporous adsorptive resins, we study on the influence factors:leaking curve,uploading concentration,dosage of washing,uploading rate, adsorption time, eluent concentration,dosage of eluent,rate of eluent,to determine the bset extraction technology of PMC.Using the technology of solid dispersion to prepare dropping pill.We study on the pharmaceutical technology of dropping pill of PMC sedative-hypnotic effect parts influence by single factor method:kind of matrix,proportion of drug and matrix,selection of condensing agent,selection of melting temperature,rate of drop,selection of dropping distance,and and establish the quality evaluation.Results: The best extraction technology of PMC was: Add 8 times the amount of 50% of ethanol solution, extracted three times, each time 1h; The purification technology of PMC total anthraquinones flavonoids: HPD600 macroporous adsorptive resins by preconditioning,concentration of original sample is 0.4g/ml used sample solution 1BV,the uploading current velocity is 2ml/min,absorbed 2h,then washed with 6BV distilled water, than used 8BV 70% ethanol as eluent, the rate of elution was 1.0mL/min.PMC dropping pill of prescription was make-up:drug/matrix=1:2 PEG4000/PEG6000 =1:1 as matrix, the melting temperature is 85℃,the eondensate is silicon oil,the dropping distance is 4cm. Conclusions: The content of the study related to the extraction technology of compound Chinese herbal, purification, the preparation technology of dropping pill and establish the quality evaluation of dropping pill of PMC sedative-hypnotic effect parts ,the results showed that the preparation achieved the desired objective, had sedative hypnotic action. |
