Di Huang Yin Zi is firstly recorded in "Huang Di Su Wen Xuan Ming Lun Fang". It is used for brain diseases especially the Alzheimer's disease (AD). But the bioactive constituents of Di Huang Yin Zi are still unclear. So we developed the the method of serum pharmacochemistry that the constituents in serum and cerebrospinal fluid after oral administration of Di Huang Yin Zi were analyzed. The constituents absorbed and their metabolites were identified preliminary.This study is aimed to develop a method for screening the bioactive constituents in serum and cerebrospinal fluid after oral administration of Di Huang Yin Zi.First of all, the fingerprints of Di Hang Yin Zi were performed by Ultra Performance Liquid Chromatography coupled with electrospray ionization (ESI) Mass spectrometry and Gas chromatography-Mass spectrometry. The common peaks of the fingerprint were identified. Secondly, using serum pharmacochemistry method, the chromatograms of rabbit serum samples after oral administration of Di Huang Yin Zi and crude drugs were analyzed and compared with control rabbit serum. Ten constituents absorbed into the blood after administration of Di Huang Yin Zi were detected, including eight metabolites and two original form compounds. According to the standard compound, one constituent was identified as Schizandrol A. Finally, constituents absorbed into rabbit cerebrospinal fluid after oral administration of Di Huang Yin Zi were analyzed and identified by Ultra Performance Liquid Chromatography coupled with electrospray ionization (ESI) Mass spectrometry and Gas chromatography-Mass spectrometry. As a result, five compounds including one original form compound and four metabolites were observed from a comprehensive analysis of the chromatography of Di Huang Yin Zi, controlled cerebrospinal fluid and dosed cerebrospinal fluid. We found the five constituents absorbed into the erebrospinal fluid matched to the compounds absorbed into the blood. According to the results, we discovered that the Schisandra chinensis made a maximal contribution to the constituents absorbed into cerebrospinal fluid. Thus, Schisandra chinensis was proved to play the most important role in the treatment of Di Huang Yin Zi for AD.This study developed a method for screening the bioactive constituents in plasma and cerebrospinal fluid after oral adminstration of Chinese herbal medicine and provided helpful chemical information for further pharmacology...
