| Background:Knee osteoarthritis (Osteoarthritis OA, knee) is one of common and multiple chronic bone and joint disease in the elderly.It is difficult to treat.It is characterized as articular cartilage degeneration, bone hyperplasia and Knee joint stiffness. Population of 38-60 years of age in 78.5% of people with osteoarthritis was one of the major decline in the quality of life. The treatment goal is to control pain and improve joint function and quality of life, inhibiting the disease process and the various side effects.Purpose:To observe the impact of mouse bone marrow mesenchymal stem cells on gene expression profile of cartilage differentiation by the efficacy serum of Bushen Yixue Water. It provide a new way on prevention and treatment of osteoarthritis of the knee. Methods:1. Select Clean-type mouse of Kunming about 18-20g of each.Conversion method based on surface area, according to 7.33g crude drug/kg orally.After 1 time per day for 7 days taking the eyeballs blood to preparation efficacy serum. It was kept in low temperature preservation.2. Select Clean-type mouse of Kunming, bone marrow stromal cells were isolated from the femur and tibia marrow by using of adherent culture.Bone marrow stromal cells were used for culture and proliferation.3. F1 generation of cells were taken into cartilage differentiation experiments, divided into control group and experimental group.Control group: still the original culture medium; experimental groups:in the original medium with Bushen Yigan Water efficacy of serum, directed into chondrocytes induced differentiation. Detect collagenâ…¡and SOX9 of mRNA expression.by RT-PCR after 5 days.Results:The cells were collected by RT-PCR amplification.The experimental group collagen II and SOX9 in the mRNA expression significantly increased compared with the control group.Conclusion:The efficacy serum of Bushen Yixue Water can induce bone marrow stromal cells into the differentiation of cartilage. |
