| Purpose:To study the inhibitory effect of Duhuojishengtang on the primary and secondary arthritis in.Adjuvant arthritis rats, and explore the mechanism of its anti-inflammatory and anti-rheumatism actionsMaterial and method:The Adjuvant arthritis rat model induced by Freund's complete adjuvant was established. To investigate the inhibitory effect of Duhuojishengtang on the primary and secondary arthritis in Adjuvant arthritis rats, the swollen volume of rats'claws were measured before and after injection of 0.1 ml FCA. The swollen volume was caculated by the water volume draining off. The test rats of were executed after injection of FCA for 28 days, sheared the hind limb above 15 mm of ankle, and made the pathological sections. The histological changes were observed by the lightmicroscop. The level of IL-1βand TNF-αin the inflammatory tissue of AA rats was determined by ELISA. The mechanism of its anti-inflammatory and anti-rheumatism actions was educed through the changes of inflammatory factors.Results:1. Duhuojishengtang with low, middle and high dose can suppress the primary swollen of AA rats'claw differently after injection of FCA for 12h. Comparing with the rats of model group, Duhuojishengtang with middle and high dose posses significant deviation (P<0.05,0.01)2. Duhuojishengtang with low, middle and high dose can suppress the primary swollen of AA rats'claw differently, and the suppressive sffect was the most significant on the 20th days. Comparing with the rats of model group, Duhuojishengtang with low, middle and high dose posses significant deviation (P<0.05,0.01)3. The pathological sections of AA rats'ankle were observed by the lightmicroscop. Duhuojishengtang with low, middle and high dose can obviously lighten swollen of synovial membrane, decrease inflammatory cell infiltration and suppress synovial cell proliferation and fibrous tissue exudation.4. Duhuojishengtang with low, middle and high dose can decrese the level of IL-1βand TNF-αin the inflammatory tissue of AA rats after administration for 18 days. Comparing with the rats of model group, Duhuojishengtang with low, middle and high dose posses significant deviation (P<0.05,0.01)Conclusion:1. Duhuojishengtang can suppress the primary swollen of rat's claw induced by FCA, and prevent the primary arthritis in Adjuvant arthritis rats.2. Duhuojishengtang can suppress the secondary swollen of rat's claw induced by FCA, and cure the secondary arthritis in Adjuvant arthritis rats.3. Duhuojishengtang can obviously lighten the swollen of synovial membrane, decrease the mononuclear macrophage infiltration and suppress the synovial cell proliferation and fibrous tissue exudation. Duhuojishengtang can protect ankle cartilage in some degree.4. Duhuojishengtang can decrese the level of IL-1βand TNF-αin the inflammatory tissue of AA rats, which infers that Duhuojishengtang may educe its anti-inflammatory and anti-rheumatism actions by suppressing the activity of inflammatory factors. |
