| Objective:Bone loss in patients with diabetes is an issue concerned about by people. A number of studies confirm that bone loss in diabetic patients is closely related with diabetic kidney disease, but both animal or clinical studies are mostly about the older body of diabetic nephropathy. The incident age of type 2 diabetes tends to younger,studies of bone metabolism in young type 2 diabetic nephropathy are few.The study observes the bone metabolism and morphological changes in young type 2 diabetic nephropathy rats via developing young type 2 diabetes rat model with abnormal bone metabolism, and provides evidence for the clinical.Methods:52 two-month-old SD male rats, in which 19 as normal control group and other 33 were developed rat model of type 2 diabetic nephropathy by combination of unilateral nephrectomy, diet enriched sucrose and fat and intraperitoneal injection of streptozotocin(STZ) for low dose.After the modeling,14 were killed at 5 month old and other 19 were killed at 6 month old.Serum calcium, phosphorus, alkaline phosphatase(ALP),1,25-dihydroxy vitaminD3, parathyroid hormone and 24-hour urine calcium and phosphorus were measured respectively.Bone mineral density (BMD) of left femur was measured.The 1/3 proximal of left tibia was used to do bone histomorphometry analysis.Results:The blood glucose of rats in model group reached in the modeling standard,24-hour urinary albumin increased significantly, and the renal pathology changing was clear,so the model of type 2 diabetic nephropathy was made successfully.Serum calcium and phosphorus levels were not significantly different during three groups. ALP,24-hour urine calcium and 24-hour urine phosphorus in the model group were significantly higher than the normal control group, no significant difference between the two later groups.The level of 1,25-dihydroxy vitaminD3 in the 6-month-group was significantly lower than the normal control group and the 5-month-group.The level of parathyroid hormone in the 6-month-group was significantly higher than the normal control group and the 5-month-group.The level of BMD in the model group was significantly decreased compared with normal control group. Indicators of trabecular bone histomorphometry were significantly diffefent compared with normal control group which showed a bone mass loss in the model group.Conclusion:The model of type 2 diabetic nephropathy was made successfully by combination of unilateral nephrectomy,sucrose and fat enriched diet and intra-peritoneal injection of streptozotocin(STZ) for low dose.For young type 2 diabetic nephropathy rats,serum calcium and phosphorus can be normal at the early period althoug so much calcium was lost though the urin,the level of parathyroid hormone and alkaline phosphatase increased significantly, The level of 1,25-dihydroxy vitaminD3 decreased significantly.The BMD and bone histomorphometry indicators chaging showed a significant bone loss for the young type 2 diabetic nephropathy group. |
