Radix P. Rubra included by China pharmacopoeia 2010 is dry P. lactiflora Pall.and P. veitchii Lynch root.It can clear away heat, cool blood, scatter stasis, relieve pain and is frequently used by traditional Chinese medicine. Among them, P. veitchii Lynch. is a wild resources, P. lactiflora Pall has wild product and cultivation product, and the latter is rich in resources, however, there is a tradition that only wild P. lactiflora Pall and P. veitchii Lynch. can be used as Radix P. Rubra. As circumstances change, and other human factors, wild resources has been badly damaged, instead,cultivation industry has constantly developed, market survey found phenomenon that dry cultivation product root was used as Radix P. Rubra has made more common. there are some differences between wild P. lactiflora Pall.and cultivation product in regional distribution, growing environment, chemical composition proportion and content, etc, how is the commodity medicinal material situation of Radix P. Rubra?is there scientific basis for that cultivation product was used for Radix P. Rubra? how to scientifically evaluate the quality of both? these need further research. This paper first makes a systematic commodity medicinal material investigation on Radix P. Rubra,and then makes a systematic comparative study on wild P. lactiflora Pall.and cultivation product, provides scientific basis for expanding medicinal materials resources and that cultivation product was used for Radix P. Rubra, has important significance of protecting the ecological resources, and promoting the sustainable development of traditional Chinese medicine. The research content as follows:1. The commodity medicinal material investigationArticle take the field investigation, telephone consultation with different methods analyzes main medicinal material distribution area of Radix P. Rubra and other resources situation, collects medicinal material 64 batch.Radix P. Rubra medicinal resources has widely distributed,29 batch is P. veitchii Lynch,12 batch is wild P. lactiflora Pall and 23 batch is cultivation product among the identified 64 batch medicinal material collected, Inner Mongolia, Hebei, Jilin, Sichuan, Anhui and Zhejiang provinces and most national other species all grow it. But wild resources has destroyed seriously, resource reserves has declined,and calendar year inventory has basically been eaten, have known as the authentic medicinal materials of inner duolun wild resources is already in productive endangered, this agreement with previous reports in the literature. Instead, cultivation industry has constantly developed, Sichuan ZhongJiang, QuXian, Anhui Bozhou, Zhejiang Hangzhou have established successively GAP base, according to traditional partition way definition(Species and production combination definition), cultivation product have a quite part can be used as Radix P. Rubra, and yield have increasing year by year.In the Market, Radix P. Rubra species is complex, use is chaos, a lot of cultivation product was saled as Radix P. Rubra, P. veitchii Lynch., wild P. lactiflora Pall and cultivation product all have become the three main commodity medicinal material source.The three main source medicinal materials quality all conforms to the pharmacopoeial standard,department bureau standard, prices has increased slowly, sales channel Displays diversity, there appear a large number of phenomenon of cultivation product which was used as Radix P. Rubra, and which already make commodity medicinal status changed about Radix P. Rubra, this section is discovered firstly. 2. The characters and microscopic characteristics comparative studyCharacters and microscopic characteristics comparison is one of the important qualitative indexes for Chinese medicinal material internal quality control, this paper studies comparatively on the appearance of characters, organelles and its contents between wild P. lactiflora Pall.and cultivation product, key-point compares their form size of parenchyma cells, cluster crystal, conduits, starch grains, cork cells, FangJing.The results show that their appearance of characters, organelles and its contents have significant differences.3. Active ingredients comparative studyThrough HPLC, Article determines quantitatively total glucosides of paeony(TGP), paeoniflorin(PAE),1,2,3,4,6-gallic acyl-D-glucose (PGG), benzoic acid and catechin among wild P. lactiflora Pall.and cultivation product, and studies on their differences and generality of chemical composition. Results show that:they mainly chemical composition is same, and the active ingredient content range also has cross, provides certain scientific basis for that dry cultivation product root was used as Radix P. Rubra.However, Mann-Whitney U test shows that the contents of PGG,PAE, TPG, total phenol, tannin, and benzoic acid are significant difference (P<0.05) between 12 batche wild P. lactiflora Pall.and 23 batch cultivation product.And from the average content, contents of TPG, Tannins, total polyphenol and benzoic acid in cultivation product are far higher than wild product,except PAE and PGG. Therefore, there are certain differences between wild P. lactiflora Pall.and cultivation product.4. Fingerprint chromatogram comparative studyThis experiment uses 12 batche wild P. lactiflora Pall.and 23 batch cultivation product as objects, studies on HPLC fingerprint chromatogram of Radix P. Rubra. First it establishes medicinal material fingerprint detection method, chromatographic column:Comatex C18 (250mm X 4.6mm,5μm);mobile phase:acetonitrile-0.1% phosphoric acid aqueous gradient elution; record:54min chromatogram; detected wavelength:230nm; column temperature:30℃; velocity: 1.0mL/min;Enter the sample weight:10μL.Second,by TCM chromatographic fingerprint similarity evaluation system (2004A edition) released by state pharmacopoeia committee,it analysis the fingerprint chromatogram of 12 batch wild P. lactiflora Pall.and 23 batch cultivation product, calibrates 13 common peak of chromatographic peak, extracts its common peak generate their both standard chromatograms. Finally, this experiment similarity evaluates the generated standard chromatograms, The results found that wild P. lactiflora Pall.and cultivation product standard fingerprint similarity between for 0.885<0.9, Show there exist certain differences among their quality, using fingerprint technology can better reflect the difference between their quality.5. Preliminary pharmacodynamic the comparative studyThrough xylene-induced mouse ear swelling test, mouse capillary permeability increased induced by acetic acid test, mouse writhing test, this paper makes a function comparison of wild P. lactiflora Pall.and cultivation product. Experiments show that:both have anti-inflammatory and analgesic action and wild product anti acute inflammation is more effective than cultivation product. However, this experiment medicine is water frying liquid, is a complex solution which contains a variety of ingredients, which kind of component is major effective component, how much is each component,we do not know,it needs to be discussed further.
