| Objective 1.To approach the relationship between T help cells 17 and Interleukin-17 of the peripheral blood and the myasthenia gravis;2.To indentify the function of T help cells 17 in the myasthenia gravis.Methods 1.To select 26 cases of MG patients, and divided into 2 groups: 12 cases in ocular myasthenia gravis(OMG) groups and 16 cases in generalized myasthenia gravis (GMG) groups; Another set of 10 cases of healthy control groups. All MG patients underwent QMG examinations.2.The 4ml blood samples from three groups anticoagulation with EDTA methods were used to research.2ml freash blood of the sample were used to analyse the peripheral blood mononuclear cell in 3 hours; the remaining serum samples were collected after centrifugation by 1000rpm×10 minutes, then stored frozen-20℃.3.Applicate direct immunofluorescence to label the anti-CD4 antibody and IL-17 A label with PE, then use Flow cytometry to examine the level of Th17 cells.4.Applicate Enzyme-linked immnosoborbent assay(ELISA)to test IL-17 concentration.5.Statistical data of the three groups were analysed by SPSS 17.0,The correlative analysis between level of Th17 and IL-17 was take in MG patients,and the correlative analysis between level of Th 17,IL-17 and the QMG, the clinical data(including the course of disease and the age of onset).Results 1.Patients in three groups compared general information, the gender difference(x2=0.619,P=0.734>0.05),age difference(t=-2.172,P=0.06>0.05) differe-nces in numbers of course of disease (t=-1.221, P=0.234> 0.05)were not statistica-lly significant.The differences in MG numbers of QMG(t=-6.550, P=0.01<0.05) were statistically significant.2. there were no significant difference of Th17 among the OMG group,GMG group and the healthy group(P=0.375>0.05).3.there were no significant difference of IL-17 among the OMG group,GMG group and the healthy group(P=0.711>0.05).4.There was significant positive correlation of Th17with IL-17 level,(7=0.879, P=0.000<0.05).5. There was no significant positive correlation of Th17 level of OMG or GMG group with course of disease, and the age of onset(P>0.05). There was no significant positive correlation of Thl7 level of OMG or GMG group with course of disease, and the age of onset(P>0.05). There was no significant positive correlation of Th17 and IL-17 level of OMG group with QMG,but There was significant positive correlation of Th17 and IL-17 level of GMG group with QMG (P>0.05).Conclusions 1.There was significant positive correlation of Thl7with IL-17 level in MG patients, It was further confirmed that Thl7 cell was new CD4+T cell characterized by secrect-ing IL-17, IL-17 is the effect factor of Th17.2.there were no significant difference of Th17 among the OMG group,GMG group and the healthy group. there were no significant difference of IL-17 between MG group and healthy control group,though the data of IL-17 of MG patients was higher than healthy control group. the result was not consistent within the EAMG.3.In our study, the result was not consistent within the EAMG. The result was not consistent within the MG, the possible causes including: referring to the document related the experimental autoimmune myasthenia gravis(EAMG),when Thl cell was remained,the function of Thl7 cells was not obvious. Thl cells maybe play an important role by cells and antibody-mediated pathway in the pathogenesis of MG, Maybe Th17 did not made decisive role. Futhermore the number of sample in our study was small, large sample need to be further confirmed.4.There was no significant positive correlation of Th17 level of MG group with course of disease, and the age of onset; There was no significant positive correlation of IL-17 level of MG group with course of disease, and the age of onset; the result demonstrated that Th17 cells were not the only factor decided the development of disease. There was no significant positive correlation of Th17 and IL-17 level of OMG group with QMG,but There was significant positive correlation of Th17 and IL-17 level of GMG group with QMG. The increased level of Th17 and IL-17 maybe reflected the severity and the disease process to some extent. |
