Objective(1)To explore the association between the expression of MK and the invasion,metastasis of choriocarcinoma by studying the expression of Midkine(MK) in human choriocarcinoma cell lines JEG-3 and JAR,invasive hydatidiform moles,hydatidiform moles and normal early pregnancy villis; (2) To explore the correlation between the expression of Midkine and characteristics of the invasion and metastasis of choriocarcinoma and to provide the evidence of Midkine inhibition and midkine antisense(MK ASODN) to control the invasion and metastasis of choriocarcinoma by comparing the expression differences of MK in human choriocarcinoma cell lines JEG-3,JAR and in normal early pregnancy villis.Methods (1)Matrigel invasion assays were used to show the difference of vitro invasion ability JEG-3 cells and JAR cells;(2)The expression of MK protein were detected by immunohistochemistry (S-P) in nomal pregnancy villis (20 cases),hydatidiform moles(35 cases),invasive hydatidiform moles (18 cases),choriocarcinoma (12cases); (3)Expression of MK protein in the human chorionic villi tissues and choriocarcinoma cell lines (JEG-3 cells and JAR cells)were detected by western blot.Results(1)The invasive cell number was significantly larger in JEG-3 cells (182106.839)than in JAR cells(100602.651),(P<0.05);(2)MK protein mainly located in cytoplasm and cell membrane of the nomal pregnancy villis,hydatidiform moles,invasive hydatidiform moles and choriocarcinoma by immunocytochemistry;And positive rate of MK in normal pregnancy villis,hydatidiform moles,invasive hydatidiform moles and chorionic carcinoma were respectively20.0%(4/20),37.1% (13/35),72.2%(13/18,91.7%(11/12).The difference between normal pregnancy villis group and invasive hydatidiform moles group,normal pregnancy villis group and choriocarcinoma group, hydatidiform moles group and invasive hydatidiform moles group, hydatidiform moles group and choriocarcinoma group,invasive hydatidiform moles group and choriocarcinoma group were statistically significant (P=0.022, P=0.009, P=0.026, P=0.010,P=0.041). (3) Protein expression of MK was significantly stronger (P<0.05)in choriocarcinoma cell lines than in the human chorionic villitissues by western blot.(4)The data suggested that there were significantly positive correlations between MK and the invasiveness of choriocarcinoma cells.Conclusion(1)MK protein mainly located in cytoplasm and cell membrane of the trophoblastic cells; (2)The expression of MK in normal pregnancy villis,hydatidiform moles,invasive hydatidiform moles and choriocarcinoma was gradually increased,and it indicates that MK may be play a role in the progress of malignant potentiality of nutrient cell; (3) Activation of MK enhance the invasion capability of choriocarcinoma.
