Objective:To observe the effect of Resveratol on the expression of Interleukin-1β(IL-1β)and Inducible nitric oxide synthase (iNOS) in Articular Chondrocyte in Experimental OA of Rabbit. To elucidate the mechanism and probability of Resveratol in the treatment of osteoarthritis.Mehtods:Six ones (three males and three females) were randomly selected from 36 New Zealand white rabbits (6-8 months old,1.5-2.5kg, 18 males and 18 females) as the control group (group A). The other 30 ones undergone Hulth technique and the model of osteoarthritis (OA) were expected to be established.8 weeks after the surgery, the successful establishment of OA model was proved by X-ray and pathology examination of six randomly selected white rabbits from the surgery ones. Then the other 24 surgery rabbits were randomly devided into four groups:model control group (group B), high dosage Resveratrol intervention group (group C), middle dosage Resveratrol intervention group (group D) and low dosage Resveratrol intervention group (group E).Then rabbits in group A and B received daily stomach irritation with Distilled water. Rabbits in group C,D and E received daily stomach irritation with Resveratrol solution (60mg/ml) in different dosage (120mg/kg/d,60mg/kg/d and 30mg/kg/d respectively) for 6 weeks. Then the rabbits were sacrificed and lateral cartilage sections of right femoral medial condyle were taken and reserved under Liquid nitrogen refrigerated condition for further examination. HE staining was administered to observe the histological morphology. The pathological changes of articluar cartilage were graded by Mankln's method under the microscope on the way of double blind. Proteins IL-1βand iNOS were analyzed by western blot. Statistical analysis was done via SPSS 17.0 software.Result:①Compared to normal control group (A group), model control group (B group) presented obvious degeneration changes under light microscope with HE staining and possessed a remarkably higher Mankin's grade (7.72 vs 0.00); compared to B group, high dosage Resveratrol intervention group (group C), middle dosage Resveratrol intervention group (group D) and low dosage Resveratrol intervention group (group E) presented lower Mankin's grades (2.42,2.63,3.13 vs 7.72) which have statistical significance (P<0.05).②IL-1(3 and iNOS expressed in cartilage tissue from all five groups; compared to that in normal control group (A group), IL-1βand iNOS in model control group (B group) increase 60.4% and 84.7% respectively, which possessed statistical significance (P<0.01).③Compared to model control group (B group), concentration of IL-1βin cartilage tissue from high dosage Resveratrol intervention group (group C), middle dosage Resveratrol intervention group (group D) and low dosage Resveratrol intervention group (group E) decreased 23.5%,24.7%and 20.3%respectively and concentration of iNOS decreased 19.8%,20.0%and 10.6%respectively, which had statistical significance (P<0.01); Compared to normal control group (A group), IL-1βincreased 22.7%,20.8%and 28.0%respectively and iNOS increased 48.1%,47.8%and 65.2%respectively, which had statistical significance (P<0.01).④Comparison of high and middle dosage Resveratrol intervention group (group C and D) presented no statistical sinificance (P>0.05). Comparison of high and middle dosage Resveratrol intervention group (group C and D) with low dosage Resveratrol intervention group (group E) presented remarkable statistical sinificance (P<0.01).Conclusion:1. Pathological examinations of the specimens have proved that OA model can be achieved from Hulth's method.2. Protein examinations of the cartilage tissue indicate that Resveratrol can effectively reduce the production of inflammatory cytokines -- IL-1βand iNOS. This dose-related effectiveness is joint-protective and provides scientific evidence for the promising therapeutic role of Resveratrol in osteoarthritis. |
