BACKGROUND:Essential hypertension (EH) and its complications are the leading cause of death in cardiovascular disease. The vascular endothelium plays an important role of the homeostasis of cardiovascular system. Nitric oxide (NO) is one of the most important mediators that are involved in the maintainance of vascular endothelium function. Asymmetric dimethylarginine (ADMA), an endogenous inhibitor of nitric oxide synthase, have been known as a cardiovascular risk factor. Intracellular ADMA generated mainly by PRMT1, suggesting that PRMT1 maybe associated with the essential hypertension. This study was designed to evaluate whether genetic polymorphism in PRMT1 is associated with hypertension susceptibility by a case-control study.METHODS:The PRMT1rs975484 polymorphism was genotyped in 221 essential hypertension patients and 220 age and gender match controls. All participants were recruitded for physical examination, questionnaire, and collected 5ml fasting venous blood. The blood were treated with phenol-chloroform extraction of genomic DNA. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to genotype the canddate single nucleotide polymorphisms (SNPs). The plasma ADMA levels were determined by ELISA and examing all the serum biochemical indicators.RESULTS:1. Significant difference in systolic blood pressure (P<0.05), diastolic blood pressure (P<0.05), fasting glucose levels (P<0.05), LDL-C (P<0.05), TC (P<0.05), Family history of hypertension (P<0.05) between cases and controls.2. Genotype frequencies for PRMT1 rs975484 CC, CG, GG genotype were 148 (67.0 %),56 (25.3 %),17 (7.7 %), respectively, in cases, and 165 (75.0 %),43 (19.5 %),12 (5.5 %), respetively, in controls. No difference in genotype distribution of rs975484 polymorphism (P>0.05).3. Significant difference in plasma ADMA levels between cases and controls (P<0.05). No difference in plasma ADMA levels among the genotypes.Conclusion:1. In Hunan Han population, PRMT1 gene rs975484CG polymorphism may not associated be with the susceptibility of essential hypertension.2. The plasma ADMA levels in essential hypertension patients were higher then the health population.
