Objective To analyse the learning memory ability and the antioxidant stress level of AD model of rats brain through establishing animal models and adopting Kidney-reinforcing and Blood-activating therapy method,Kidney-reinforcing therapy method,Blood-activating therapy method, at the same time compare the better Chinese medical therapy.Methods 60 SD male rats were enrolled in the study, and they were randomly divided into six groups:blank controls group, sham controls group, model group, Kidney-reinforcing and Blood-activating therapy group, Kidney-reinforcing therapy group, Blood-activating therapy group。The rat model of AD was established injecting amyloid-Aβ25-35. 21 st days after operation it was tested whether or not Morris water maze behavior would be successful.After Morris water maze behavior's test the rats in three Chinese medicine groups were irrigated into their stomaches with different Chinese medicine Kidney-reinforcing and Blood-activating, while the other three groups irrigated by physiclogical saline once a day for eight weeks. Afer that carry out Morris water maze behavior detection again. Later put them to death and took out hippocampal organizations and forehead cortex which received Trxl immunohistochemical experiments and Trx2 mRNA RT-PCR experiments.Results 1. AD model was established successfully:Free preclinical of rats in the model group and three Chinese medicine groups was obviously longer than that of normal group (p<0.01), the time staying in the original quadrant and times crossing original platform became less obviously (p<0.05).2. After traditional Chinese medicine intervention, latency or space exploration ability was improved obviously (p<0.05). Rats in Kidney-reinforcing and Blood-activating group had better space study memory ability than those in Kidney-reinforcing group and Blood-activating group. 3.Hippocampus Trxl and forehead cortex tissue Trx2 in three Chinese medical groups increased obviously (p<0.05) and those in Kidney-reinforcing and Blood-activating group were higher than Kidney-reinforcing group and Blood-activating group.Conclusion 1. The results of the study show AD model can be established successfully through amyloid-Aβ325-35 sterotaxis injection into the bilateral Hippocampus.2. Kidney-reinforcing and Blood-activating therapy method,Kidney-reinforcing therapy method,Blood-activating therapy method all can improve the rats'of AD model learning memory ability and the treatment of Kidney-reinforcing and Blood-activating therapy method is better than Kidney-reinforcing therapy method or Blood-activating therapy method.3. Kidney-reinforcing and Blood-activating therapy method,Kidney-reinforcing therapy method,Blood-activating therapy method can improve the antioxidant stress level of AD model of rats'brain,and Blood-activating Kidney-reinforcing and Blood-activating is better than Kidney-reinforcing therapy method or Blood-activating method. |
