Delila(del), roseal(ros),which are the regulatory gene of the anthocyanins cloned fromsnapdragon,are respectively regulated by the the fruit-specific promoter PS and transformcommonly strawberry to improve the content of anthocyanins and expect to change fruits color.1. On the base of extracting of goldfish grass total RNA,designing of primer according tothe del,ros gene sequences logged on NCBI, del,ros were cloned.It is showed that the similarityof the genetic sequence of cloning and the gene bank in the world are more than98%.2. On the base of adding the righe extinction enzymes enzyme cut site and designingprimer,fruit specific promoter PS is getted from plasmid pCAMBIA3300-PS-RS-Tnos which iskeeped in Lin Zhongping laboratory in Beijing university.3. Constructing the middle expression vector: pBI121-PS-del-Tnos,pCAMBIA1301-PS-ros-Tnos on the baise of that del,ros are respectively regulated by the thefruit specific promoter PS,by which construcing the plant expression vector:pCAMBIA2301-PS-del-Tnos-PS-ros-nos.4. The plant expression vector is transformated into the tumefaciens AGL0,and thentransform strawberry by agrobacterium mediated dordrecht,obtain resistant plant by screeningwith nptⅡ.
