In this paper,He-Ne laser irradiation enhanced the role of protein in leaves of wheat after UV-B Radiation damage Repair two-dimensional electrophoresis. Wheat(Jin mai NO.79) leaves as materials, setting the normal light group (CK),enhanced UV-B radiation group (B) and the UV-B and laser composite treated(LB).The wheat seed containing Petri dish with wet filter paper was cultured at25℃, Wheat seedling after germination by10.08kJ·M2·D1enhanced UV-B radiation,then10mW·mm-2He-Ne laser is irradiated.By measuring total protein in wheat seedling leaves, Using two-dimensional electrophoresis technology, differences in protein profiles obtained by PDquest8.0software analysis, looking for differences in protein spots before and after radiation. Analytical results of the role of He-Ne laser damage repair of UV-B on wheat leaf protein showed that: (1)Wheat leaves after six days by the UV-B radiation, plants obviously dwarf, curled leaf, radicle curved upward. He-Ne laser treatment, the wheat plant height, leaf, radicle curved markedly reduced.(2)From the experimental results of He-Ne laser radiation, enhanced enzyme activity in wheat leaves after UV-B radiation, the synthesis of proteins.(3)Through the control of the normal light group,and the UV-B radiation treated wheat leaf protein profiles were found in21point difference, including6unregulated proteins,12proteins down-regulated genes, explicitly expressed in points3.Control the group of UV-B radiation treatment and UV-B and laser composite treatment groups were found24differentially expressed proteins,including14unregulated proteins,6proteins down-regulated genes, specifically expressed in points4. Some of the differentially expressed proteins by mass spectrometry, three protein spots were identified.
