Wheat yellow dwarf disease, a kind of serious virus disease transmitted by aphids, often causes poor growth, serious dwarf, extremely low leaf-blight earing rate and even little production in wheat. At present, prevention of the disease is still restricted to aphids control using chemical pesticides, sowing time adjustment based on migration period of aphids and hybrid breeding resistant varieties. However, these strategies is either environmentally hazardous or time and resources consuming with unsatisfactory results. With the development of molecular biology, transgenic technology is more and more applied to plant resistance breeding. On the basis of the previous work by other researchers, we investigated disease resistance, genetic stability and biological safety of the transgenic strains. The results are stated here:(1) By PCR and PCR-southern blot assay, we investigated genetic stability of genetically modified Yang wheat strain T4 generation, which has been introduced a successfully RNAi vector constructed with CP gene of barley yellow dwarf virus-GAV. Data shows that T4 generation transgenic Yang wheat strain is stable genetic.(2) Transgenic wheat strains express obviously more resistance to BYDV-GAV and better agronomic traits than non-modified varieties and the original varieties Yang wheat 158, however, different transgenic plant show different level of resistance. The results predict that different resistance of each transgenic T4 generation plant may caused by copy number of BYDV and the influence of chromosome separation chain.(3) We found that T4 generation of transgenic wheat have no effect on fungal disease and field biological weeds and it is not been observed that structure variations of the field biological community by the related biological safety survey for T4 generation transgenic Yang wheat strain in the field. Genetically modified organisms also need more research and discussion of biological safety by the method of combination with field experiments and laboratory experiments in a long time.(4) Conventional crossbreeding has been applied for T4 generation genetically modified wheat with three commercial varieties of xiaoyan 6, xiaoyan 22 and xinong 979 by the method that combined with reciprocal cross, then it is obtained that part of the F1 generation hybrid wheat, which provided test materials for next step to continue the experiment.
