In order to study the characteristics of fish collagen and extraction conditions,in this experiment,the skin and swim bladder of bighead carp as raw materials to extract low molecular weight collagen peptides by enzymatic hydrolysis Enzyme,and based on physical characteristics of the collagen, select the optimum extraction conditions of collagen using single factor experiments ,the optimum extraction conditions of skin collagen is :pH is 4,reaction temperature is 30℃,substrate concentration 30g/L~40g/L,the concentration of enzyme to substrare ratio is 1500u/g,reaction time is 5h ;the optimum extraction conditions of swim bladder collagen is :pH is 4.8~5,reaction temperature is 30℃,substrate concentration 40g/L,the concentration of enzyme to substrare ratio is 2000u/g,reaction time is 5h .The extraction rate of skin collagen can be reached 27.26%,and the rate of swim bladder is achieved 16.73%. The smaller the molecular weight of collagen to be absorbed more easily, the collagen protein whitch extracted by enzymatic hydrolysis mixed acetic acid soaking, then the molecular chain is chopped to peptides, the molecular weight of collagen can be controlled about 3000D. we can filter out the melanoma in skin collagen by the 3mm diameter microporous membrane,so that the colour of collagen becomes white,in addition,we have done routine analysis on water,ash,fat,protein and amino acids of skin and swim bladder. Determined the denaturation temperature of the skin and swim bladder collagen.are both 30℃.
