This paper studies the combination of water and enzyme to solvent extract rice bran oil and researches the effects on the extraction productivity from different enzymes. After confirming the collocation of enzymes, optimized the technology of combination of water and enzyme to solvent extract rice bran oil through single factor experiments and response surface experiments. The physical and chemical properties of rice bran oil were compared the optimized technology with use organic solvent directly adopts anti-phase extraction, including:acid value, iodine value, peroxide value, unsaponification content and FTIR and fatty acid analysis of two kinds of oil. We also studied the effects of rice bran protein productivity and purity on pH of alkali and washing times, compared the physical and chemical properties of rice bran protein by three methods of alkali extraction, joined papain extraction and use cellulose enzyme zymolysis then to alkali extrsction, including:solubility, hygroscopicity and moisturizing, emulsification and foaming ability. We studied on the main technical parameters of the microcapsules which were prepared by rice bran oil, and determined this product. Finally, experimental conclusions as follows:1) When using pectinase, cellulase and papain respectively extracted rice bran oil, extracting by cellulase had good result. The extraction rate is 11.46%. When using two from three enzymes extracted rice bran oil, it had best result by papain and cellulose, its extraction rate is 13.12%. Extraction rate by using three enzymes together is very similar with using papain and cellulase. Therefore, extracting rice bran oil is used with papain and cellulase.2)We use Design Expert (7.0) software to do the response surface optimization experiments of four factors by three levels that the ratio of liquid and materials, time of enzymatic hydrolysis, addition amount of cellulose enzyme, addition amount of papain on the basis of single factor experiments, it is concluded that the optimal conditions of extracting of rice bran oil for:the ratio of materialand liquid was 1:6,time of enzymatic hydrolysis was 2.Oh, addition amount of cellulose enzyme was 0.38%, addition amount of papain was 0.23%. In these conditions, extraction productivity of rice bran oil was 13.45%.3) Relevant indicators of solvent extracting rice bran oil by the combination of water and enzymes, is better than the indicators of reverse extracting rice bran oil by organic solvent.4) Compared alkaline extracting rice bran oil with using papain extraction and using cellulase and alkaline extraction, found using cellulase and alkaline extraction is the best way.5) On the studies of physical and chemical properties of protein, we found that the solubility, hygroscopicity and moisturizing property were better of rice bran protein extracted by papain, protein emulsification and emulsion stability, foaming ability and stability have great relevance with protein solubility.6) According to emulsion stability, viscosity and product performance of micro-capsules, it is determined that preparation of rice bran oil micro-capsules needs 20Mpa homogenization pressure, the average size of emulsion is 398.4nm, solids concentration is 40%, spray drying inlet air temperature is 190℃, the air temperature is 95℃; Embedding rate of rice bran oil micro-capsules is 80.30%, moisture content is 2.49%, melt solution temperature (Tm) is 142.88℃, heat enthalpy is 9.7809J/g; oven storage accelerated oxidation experiments show that rice bran oil and its micro-capsules from Induction period to divergent period needs about 20 days, and the storage time that rice bran oil micro-capsules reached standards POV values is 1.35 times than the ones that rice bran oil reached standards POV values.
