PCR is Polymerase Chain Reaction, a technology of DNA expansion in vitro, which is an import technology in molecular biology. To the system of PCR gel image, correct qualitative and quantitative to PCR gel image are obtained only after correcting analysis and extraction for it. This system mainly includes that image denoising, image enhancement, the division of lanes and bands, image segmentation, optical density (OD) value of band to PCR gel image. Low-pass filtering is used to denoise the image. Top and low hat is used to enhancement the image. Gaussian smoothing is used to vertical projection curved line of image in the division of lanes and bands, through the smooth line to get the threshold value of the division of lanes and bands and divise the lanes and bands of PCR gel image. Adaptive of global threshold is used to segment the image. The Gray Intensity of image is used to count the optical density (OD) value of image in the last. When we get the value of integral optical density, Lambert-Beer Law is used to get the quantitative of PCR gel image. Test sample is compare to standard sample, the correct qualitative of the PCR gel image can be obtained. The final test results showed that the time of dividing bands and lanes were reduced, and the precision of dividing and final quantitative analysis were improved.
