Purpose: To study the chemical compositions of the fruits of Acanthopanax sessiliflorus(Ruqr.et Maxim)Seem.and develop an HPLC method for determination of the contents of hyperin and seopoletin .Material and method:Material: The fruits of Acanthopanax sessiliflorus(Ruqr.et Maxim) Seem.were taken on the outskirts of Dandong and were identified by Professor Wang Bing of Liaoning University Of Traditional Chinese Medicine.Method:1. The methods of solvent extraction, silica gel column, Sephadex LH-20 column and recrystallizition were used to the extraction and isolation of compounds; And the methods of spectroscopy were used to the structure identification.2. The contents of hyperin and scopoletin were determined by HPLC.Results:1.20 compounds were isolated from the fruits of Acanthopanax sessilifloru(sRuqr.et Maxim) Seem.And 18 of them were identified. They are characterized as [(-)sesamin](1),β-sitosterol(2),chiisanogenin(3),scopoletin(4),daucosterol palmitate(5),daucosterol(6),stigmasterol(7),protocatechuic acid(8),scopolin(9),quercetin(10),hyperin(11),oleanolic acid 3-o-6'-methyl-β-D-glucuronopyranoside(12), succinate(13),oleanolic acid 3-o-6'-buthyl-β-D-glucuronopyranoside(14),momordin lb(15),oleanolic acid(16),3-O-[(α-L-arabinopyranosyl)-(12)]-[-D-glucuronopyranosyl-6- Omethyl ester]-olean-12- ene-28-olic acid(17),galactitol(18),2.EstabLishing the content determination method of fruits of Acanthopanax sessiliflorus(Ruqr.et Maxim) Seem.by HPLC with gallic acid as determination. The method is simpLe, accurate, reproducible . The liner ranges of hyperin and seopoletin were 0.06912-0.3456μg(r=0.9999)and 0.01424-0.09968μg(r=0.9999),respectively. The recoveries of hyperin and seopoletin were 100.9%and99.6%. Conclusions:1.Isolated compounds from Acanthopanax sessiliflorus (Rupr.et Maxim.)Seem. and identified their stuctures.Compound 4,13,14,18were firstly isolated from the fruits of Acanthopanax sessiliflorus (Rupr.et Maxim.)Seem..2.EstabLishing the content determination method of Acanthopanax sessiliflorus (Rupr.et Maxim.)Seem. .The method is accurate and reliable that can be used to the quality control of fruits of Acanthopanax sessiliflorus (Rupr.et Maxim.)Seem..
